Subbing slides.

CSH Protoc

Published: September 2008

INTRODUCTIONWhen mounting sections on a slide, it is essential that the slide be ultraclean. To ensure that the section sticks to the slide, slides can be covered with a thin layer of gelatin. The process of cleaning and gelatin-coating is called "subbing." Several boxes of slides should be subbed at a time and stored, covered, in the refrigerator until they are needed. Subbed slides can also be purchased commercially.

This unit covers some basic procedures that are common to a wide range of neuroanatomical protocols for brain tissue. Procedures are provided for preparation of unfixed fresh brain tissue as well as for perfusion fixation of animals to obtain fixed neural tissue. A variety of methods for sectioning are described, including frozen sectioning using a cryostat or microtome and sectioning with a vibratome.

INTRODUCTIONIt is imperative that the slides and coverslips used in fluorescence microscopy procedures be extremely clean. Although coverslips look clean, especially when a new box is first opened, they may have a thin film of grease on them that will not allow tissue culture cells to adhere well and that may interfere with some processing steps in certain protocols. Therefore, coverslips should routinely be washed with acid or base solutions to rid them of this film.
