Published: December 2011

A novel species, Aspergillus saccharolyticus sp. nov., belonging to the Aspergillus section Nigri group is described. This species was isolated in Denmark from treated hardwood. Its taxonomic status was determined using a polyphasic taxonomic approach including phenotypic (morphology and extrolite profiles) and molecular (β-tubulin, internal transcribed spacer and calmodulin gene sequences, and universally primed PCR fingerprinting) analysis. Phenotypic and molecular data enabled this novel species to be clearly distinguished from other black aspergilli. A. saccharolyticus is a uniseriate Aspergillus species that is morphologically similar to Aspergillus japonicus and Aspergillus aculeatus, but has a totally different extrolite profile compared to any known Aspergillus species. The type strain of A. saccharolyticus sp. nov. is CBS 127449(T) (=IBT 28509(T)).

Efficient heterologous expression of cellobiose 2-epimerase gene in Escherichia coli under the control of T7 lac promoter without addition of IPTG and lactose.

Article Synopsis
  • The study focused on expressing the cellobiose 2-epimerase gene (csce) from Caldicellulosiruptor saccharolyticus in E. coli using TB medium with specific supplements, aiming to enhance enzyme activity.
  • Results showed that when IPTG and lactose were omitted, enzyme activity reached 5.88 U/mL, significantly higher than when 1.0 mM IPTG was included.
  • The findings highlight that certain supplements like yeast extract and soy peptone can induce gene expression without IPTG and lactose, leading to a 2.2-fold increase in enzyme activity, which could improve recombinant protein production strategies in E. coli.

Article Synopsis
  • * It thrives in a temperature range of 25-45 °C, prefers a neutral pH of 7.0, and can tolerate low salt concentrations (optimum at 1% NaCl).
  • * Phylogenetic analysis shows that RY-2 is closely related to other strains within its genus with a high sequence similarity to JCM 17389 (98.3%) and is proposed as a novel species with the type strain designated as RY-

Thousands of lakes harbouring different characteristics (pH, salinity, temperature) are located on the Tibetan Plateau, and the mining of microbial resources inhabited in these lakes has great value. Two Gram-stain-negative, aerobic, rod-shaped, non-motile strains (LQ15W and AIY15W) were isolated from freshwater lakes on the Tibetan Plateau. Comparisons based on the 16S rRNA gene sequences showed that both strains LQ15W and AIY15W share 16S rRNA gene sequence similarities 98.

a coagulase-negative staphylococcal species, has some unusual characteristics for human-associated staphylococci, such as slow growth and its preference for anoxic culture conditions. This species is a relatively abundant member of the human skin microbiota, but its microbiological properties, as well as the pathogenic potential, have scarcely been investigated so far, despite being occasionally isolated from different types of infections including orthopedic implant-associated infections. Here, we investigated the growth and biofilm properties of clinical isolates of and determined host cell responses.

Matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry provides accurate species-level identification of many, microorganisms retrieved from bovine milk samples. However, not all those microorganisms are pathogenic. Our study aimed to: (1) determine the species-specific prevalence of microorganisms identified in bovine milk of apparently healthy lactating quarters vs.
