The components of an HPLC system can be easily reassembled into an instrument for transient-state enzyme kinetic experiments based on the continuous-flow technique. The scheme of reassembly, operation and data evaluation is described in detail for the Pharmacia FPLC system. The working quality of the suggested scheme was tested using a well-studied process of bacterial beta-galactosidase activation by Mg2+ ions. The found parameters were in a good agreement with the literature data. The pulse-flow mode was developed for the delivery of working solutions to reduce the required quantities of enzyme and/or other reaction components. The work presents new approach to HPLC modules as building blocks for sophisticated liquid flow reactors, such as automatic solid phase synthesizers, sequencers and so on.

An easy applicable and selective sample preparation technique has been developed for trace and simultaneously analysis of Glipizide (GLP) and Pravastatin (PST) molecules in biological matrices based on magnetic solid phase extraction (MSPE) and high-performance liquid chromatography (HPLC). A new magnetic adsorbent including FeO@TEOS-Melamine has been synthetized and characterized for extraction studies. A rapid, simple, and cost-efficient extraction method was developed for evaluating and screening benzo(a)pyrene (BaP) in tea samples by using high performance liquid chromatography (HPLC) with coupled fluorescence detector (FLD) in order to obtain the best extraction performance. In this study, it was observed that when optimized using microwave assisted extraction (MAE) method was performed twice for 2 min using 10 mL n-hexane: acetonitrile (1:3, v/v). The recoveries for BaP in tea were found to be 97 ± 2; 83 ± 8 and 92 ± 6%, respectively.

Introduction: This study evaluated the relationship between total plasma and free kidney concentrations of amphotericin B (AmB) in healthy and -infected Wistar rats using microdialysis and has the potential to significantly impact future research in this field and promote the development of antifungal drugs. The findings of this study, which show that plasma levels are a good predictor for AmB kidney concentrations and can be used to optimize its dosing regimen, underscore the importance of this research.

It is well known that hydroxyl radical (OH) can be largely produced either through the classic iron-mediated inorganic-Fenton system or our recently discovered haloquinones/HO organic-Fenton-like system, but rarely produced via thiol compounds. Here, unexpectedly, we found that OH can be unequivocally generated by incubation of HO and mercaptotriazole (MTZ), a typical heterocyclic thiol which has been used as an environmentally friendly corrosion inhibitor for mild steel. By the complementary applications of HPLC-MS and oxygen-18 isotope-labeling method, MTZ-derived sulfenic (MTZ-SOH) and sulfinic acids were detected and identified as transient intermediates, and sulfonic acid as final products.
