A mathematical model for research on the refractive index profile (RIP) of multistep ion exchange processes (IEPs) of gradient refractive index rod lenses (GRINs) is established by the different initial condition and boundary condition, based on the Fickian diffusion equation. GRIN rod lenses have been fabricated using the three-step IEPs. Research results indicate that the experimental deviations of refractive index (DRI) are in good agreement with the theoretical data. The DRI of three-step IEPs is superior to the one- and two-step IEPs and smaller than 10(-5).

Background: High-precision and broad-range pH detection is critical for health status assessment, such as signal transduction, enzyme activity, endocytosis, and cell proliferation and apoptosis. Although pH-responsive ratiometric fluorescent probes offer an effective pH monitoring strategy, their preparation often requires multi-step modification and decreases fluorescence efficiency and stability. Herein, we developed a simple method to prepare fluorescent Si dots with dual emission centers for high-precision and broad-range pH monitoring, and the detection of urease based on pH-responsive Si dots and pH monitoring in living cell was further explored.

Ion exchange chromatography (IEX) is an important analytical technique for the characterization of biotechnology-derived products, such as monoclonal antibodies (mAbs) and more recently, cell and gene therapy products such as messenger ribonucleic acid (mRNA) and adeno-associated viruses (AAVs). This review paper first outlines the basic principles and separation mechanisms of IEX for charge variant separation of biotherapeutics, and examines the different elution modes based on salt or pH gradients. It then highlights several recent trends when applying IEX for the characterization of biotechnology-derived products, including: i) the effective use of pH gradients, ii) the improvement of selectivity by using organic solvents in the mobile phase, multi-step gradients, or by combining ion pairing and ion exchange, and iii) the increase in analytical throughput using ultra-short columns or automated screening of conditions.

Few studies are concerned with the effect of the conjugat protein on the bioactivities of the abalone gonad polysaccharide (AGP). In this study, a series of treatments, including raw material (female and male) defatting, extraction temperature (25-121 °C), proteolysis, ultrafiltration, and ethanol precipitation, was conducted to investigate the role of the conjugate protein on AGP anticoagulant activity. All AGP extracts significantly prolonged activated partial thromboplastin time (APTT) and thrombin time (TT).
