Background/aims: Endometriosis is a benign but chronic disorder associated with pelvic pain and infertility. Enhanced proliferation and reduced apoptosis susceptibility are characteristics of endometriosis. Cordycepin is a poly(A) polymerase inhibitor. It induces shortening of poly(A) tails, leading to destabilization of mRNAs and finally to proliferation inhibition and cell death in normal and tumor cells. The potential of cordycepin to block proliferation and survival of 11z human immortalized epithelial endometriotic cells was determined.

Methods: 11z cell cultures were treated with cordycepin. Cordycepin-induced inhibition of proliferation and alterations in protein expression and protein phosphorylation were determined by the methyl thiazolyl tetrazolium assay and immunoblot analysis, respectively.

Results: Cordycepin induced the rapid and significant upregulation of the cell cycle progression inhibitor p21 and the downregulation of the cell cycle progression promoter cyclin D(1), finally leading to the inhibition of the proliferation of 11z human epithelial endometriotic cells. Cordycepin reduced the phosphorylation of the p38 mitogen-activated protein kinase and the retinoblastoma protein. It also activated caspase-dependent, intrinsic apoptosis, as documented by the proteolytic cleavage of the caspase-9, caspase-3 and the poly(ADP ribose) polymerase 1 precursor.

Conclusion: The mRNA polyadenylation inhibitor cordycepin inhibits proliferation and survival of endometriotic cells.

Background: Endometriosis patients exhibit a cancer-like glycolytic phenotype. The pyruvate kinase M2 (PKM2)/hypoxia-inducible factor-1 alpha (HIF-1α) axis plays important roles in glycolysis-related diseases, but its role in patients with endometrial polyps (EPs) combined with endometriosis has not been validated.

Methods: EP samples were collected from patients with and without endometriosis.

Background: Nuclear receptor subfamily 4 group A member 3 () is lowly expressed in ectopic endometrium and can be degraded by ubiquitination in vascular endothelial cells. Murine double minute 2 () is predicted to be the ubiquitin ligase of . Hence, we investigated the effects of and on endometriosis and clarified corresponding regulatory mechanisms.

As a chronic gynecological disease, endometriosis is defined as the implantation of endometrial glands as well as stroma outside the uterine cavity. Proliferation is a major pathophysiology in endometriosis. Previous studies demonstrated a hormone named melatonin, which is mainly produced by the pineal gland, exerts a therapeutic impact on endometriosis.

This study investigated the effect of Luoshi Neiyi Formula(LSNYF) on hypoxia-inducible factor-1α(HIF-1α) and steroidogenic factor 1(SF-1) in endometriosis(EMs), aiming to explore the mechanism of Luoshi Neiyi Formula in treating EMs. Immunohistochemistry and quantitative real-time PCR(qPCR) were employed to determine the expression of HIF-1α and SF-1 in the endometriotic tissue. Human primary endometrial stromal cells(ESCs) were extracted and identified, in which the expression levels of HIF-1α and SF-1 were measured by immunofluorescence and qPCR.
