The tetrahydroisoquinoline skeleton is a pharmacologically significant core structure containing chiral centers, making enantiomeric separation crucial due to the potentially distinct biological effects of each enantiomer. In this study, laudanosine (-methyl-tetrahydropapaverine) and its three derivatives (6'-bromo-laudanosine, norlaudanosine, and -propyl-norlaudanosine) were synthesized and used as model compounds to investigate chiral recognition mechanisms. Screening over twenty cyclodextrins (CyDs) as chiral selectors in capillary electrophoresis (CE), we found anionic CyDs to be the most effective, with sulfated-γ-CyD (S-γ-CyD) achieving a maximum of 10.

Background: Significant methodological shortcomings have been documented to date in risk minimisation program evaluations for medicinal products, including overreliance on survey methods alone. Recently updated guidances from the Food and Drug Administration (FDA) and the European Medicines Agency (EMA) recommend the use of frameworks and mixed methods designs to improve the rigor of these assessments.

Objective: The purpose of this paper was to exemplify how a mixed methods approach, guided by an implementation science framework, can be used to design the evaluation of a risk minimisation program.

Objectives: Small interfering RNA (siRNA) can silence disease-related genes through sequence-specific RNA interference (RNAi). Cationic lipid-based liposomes effectively deliver nucleic acids into the cytoplasm but often exhibit significant toxicity. This study aims to synthesize a novel ionizable lipid, Nε-laruoyl-lysine amide (LKA), from natural amino acids, constructed LKA-based liposomes, and perform physicochemical characterization and cell-based experiments to systematically evaluate the potential of these ionizable lipid-based liposomes for nucleic acid delivery.
