The association of epithelial cord cells and ductlike structures in 2 cases of pancreatic islet tumors is presented. The first case was characterized by multiple pancreatic nodes of beta cell insulinoma in a 35-year-old woman. The second case was a Zollinger-Ellison syndrome in a 10-year-old male child with perforated duodenal ulcer due to an islet cell tumor of an aberrant pancreas in the omentrum. Stress is laid upon the striking continuity of the tumorous endocrine and exocrinelike histological formations reproducing the morphological pattern of ducto-insular relationships in histogenesis and islet regeneration.

Objectives: Head and neck malignancies (HNMs) encompass a variety of cancers that affect the oral and para-oral tissues, the most common of which are squamous cell carcinomas. Radiotherapy is commonly used to treat these cancers, often involving radiation exposure to the salivary glands. This study aims to investigate the early impacts of radiotherapy on the internal microstructure of the salivary gland cells and identify which gland exhibits the highest level of radiosensitivity.

Pleomorphic adenoma (PA) is a benign salivary gland tumor with diverse cytomorphological and architectural features, typically presenting biphasic ductal structures within a chondromyxoid matrix. We report a unique case of PA where dense lymphoid infiltration disrupted these structures, resulting in duct-like slit structures lined with a single layer of spindle cells, lacking the biphasic pattern. These spindle cells demonstrated myoepithelial nature, confirmed by positive immunostaining for pan-cytokeratin, S100 protein, and calponin.

Volumetric analysis of the terminal ductal lobular unit architecture and cell phenotypes in the human breast.

The major lactiferous ducts of the human breast branch out and end at terminal ductal lobular units (TDLUs). Despite their functional and clinical importance, the three-dimensional (3D) architecture of TDLUs has remained undetermined. Our quantitative and volumetric imaging of healthy human breast tissue demonstrates that highly branched TDLUs, which exhibit increased proliferation, are uncommon in the resting tissue regardless of donor age, parity, or hormonal contraception.

We herein report two- (2D) and three-dimensional (3D) culture methods of cholangiocytes originating from extrahepatic bile ducts of biliary atresia (BA) patients. Cells were stabilized for in vitro analyses, and 3D culture by two different methods showed the structural and functional features of cholangiocytes in the gel scaffold. First, cells were obtained from gallbladder contents or resected tissues of patients at surgery and then cultured in our original conditioned medium with a cocktail of signaling inhibitors that maintains the immaturity and amplification of cells.

Identification of Ppy-lineage cells as a novel origin of pancreatic ductal adenocarcinoma.

The Ppy gene encodes pancreatic polypeptide (PP) secreted by PP- or γ-cells, which are a subtype of endocrine cells localised mainly in the islet periphery. For a detailed characterisation of PP cells, we aimed to establish PP cell lines. To this end, we generated a mouse model harbouring the SV40 large T antigen (TAg) in the Rosa26 locus, which is expressed upon Ppy-promoter-mediated Cre-loxP recombination.
