An SAR study on the Dmt-substituted enkephalin-like tetrapeptide with a N-phenyl-N-piperidin-4-ylpropionamide moiety at the C-terminal was performed and has resulted in highly potent ligands at μ and δ opioid receptors. In general, ligands with the substitution of D-Nle(2) and halogenation of the aromatic ring of Phe(4) showed highly increased opioid activities. Ligand 6 with good biological activities in vitro demonstrated potent in vivo antihyperalgesic and antiallodynic effects in the tail-flick assay.

Astaxanthin (ASX), "king of carotenoids", is a xanthophyll carotenoid that is characterized by a distinct reddish-orange hue, procured from diverse sources including plants, microalgae, fungi, yeast, and lichens. It exhibits potent antioxidant and anti-ageing properties and has been demonstrated to mitigate ultraviolet-induced cellular and DNA damage, enhance immune system function, and improve cardiovascular diseases. Despite its broad utilization across nutraceutical, cosmetic, aquaculture, and pharmaceutical sectors, the large-scale production and application of ASX are constrained by the limited availability of natural sources, low production yields and stringent production requirements.

Thioredoxin reductase 1 (TrxR1) is an important therapeutic target for nonsmall cell lung cancer (NSCLC) treatment due to its overexpression in NSCLC cells. In this work, to address the deficiency that sesquiterpene lactone containing α-methylene-γ-lactone moiety was rapidly metabolized by endogenous nucleophiles, series of novel thioether derivatives were designed and synthesized based on a reactive oxygen species (ROS)-triggered prodrug strategy. Among them, prodrug exhibited potent cytotoxicity against NSCLC cells and better release rates in response to ROS.

Dual atomic nanozymes (DAzymes) are promising for applications in the field of tumor catalytic therapy. Here, integrating with ultrasmall FeC nanoclusters, asymmetric coordination featuring Janus Zn-Fe dual-atom sites with an ON-Fe-Zn-N moiety embedded in a carbon vacancy-engineered hollow nanobox (Janus ZnFe DAs-FeC) was elaborately developed. Theoretical calculation revealed that the synergistic effects of Zn centers acting as both adsorption and active sites, oxygen-heteroatom doping, carbon vacancy, and FeC nanoclusters jointly downshifted the d-band center of Fe 3d orbitals, optimizing the desorption behaviors of intermediates *OH, thereby significantly promoting catalytic activity.
