The effect of L-lysine HCl concentrations, pH, temperature and cultivation conditions on the synthesis of intracellular lysine decarboxylase (LDC) by Vibrio sp. was studied. The highest LDC activity was observed in a 4-hour culture grown at 30 degrees on a nutrient medium, pH 5.15 containing 0.9% L-lysine HCl as an inducer. Under these conditions the LDC activity of the bacterium was 8.43 U/mg dry cells.

Plant-Scale Circular Economy Using Biological Reuse of Electrolyte Residues in the Amino Acid Industry.

Bioengineering (Basel)

December 2024

CJ BIO Research Institute, CJ CheilJedang Corp., Suwon-si 16495, Gyeonggi-do, Republic of Korea.

The amino acid industry generates significant amounts of electrolyte residues, such as ammonium sulfate, acetic acid, and phosphoric acid, which cause challenges to sustainability. This short article investigates the feasibility of implementing a plant-scale circular economy through the recycling and biological reuse of these electrolyte residues. Scenario analyses of L-lysine (LYS) HCl, L-methionine (MET), and L-cysteine (CYS) HCl production highlight the environmental and economic benefits of the plant-scale circular economy.

Castrate-resistant prostate cancer (CRPC) is one of the most difficult cancers in men and is characterized by a poor prognosis and a high risk of metastasis. The overexpression of the disruptor of telomeric silencing 1-like (DOT1L), which is a specific methyltransferase for histone H3 at lysine residue 79 (H3K79), has been related to poor outcomes in patients with CRPC. Therefore, targeting DOT1L is considered a potential therapeutic approach to overcome the significant medical challenges of CRPC.

Background: Formulation screening is essential to experimentally balance stability and viscosity in high-concentration mAb formulations. We developed a high-throughput approach with automated sample preparation and analytical workflows to enable the integrated assessment of excipient compatibility and viscosity of mAb formulations.

Methods: Ninety-six formulations of a trastuzumab biosimilar were screened by combining 8 types of excipient modifiers with 4 types of buffers across a pH range of 4.

Article Synopsis
  • The study examined how different levels of digestible lysine (dLys) in the diet affect the productivity and health of breeding Japanese quails.
  • Results showed that there were notable improvements in egg production, egg mass, and feed efficiency with an optimal dLys level of 0.885%.
  • While increased dLys levels positively influenced certain production traits, other factors like egg quality, reproductive performance, and immune responses remained largely unaffected.

Article Synopsis
  • The study focuses on improving biopharmaceutical drug formulations by investigating various buffer systems for monoclonal antibodies (mAbs) to enhance protein stability.
  • While traditional buffers like histidine and phosphate are commonly used, the research shows that alternative buffers, particularly arginine/citrate and histidine/citrate, can significantly improve stability under stress conditions.
  • The results indicate that relying solely on conventional buffers can limit the effectiveness of biopharmaceutical formulations, as many alternatives demonstrate equal or better performance in maintaining protein integrity.
