A simple, rapid, selective and sensitive HPLC-UV method has been developed and validated for the determination of ponicidin in rat plasma. The analyte was extracted from rat plasma by liquid-liquid extraction with ethyl acetate as the extraction solvent. The LC separation was performed on a Zorbax Eclipse XDB C(18) analytical column (150 × 4.6 mm i.d., 5 µm) with an isocratic mobile phase consisting of methanol-water-phosphoric acid (45:55:0.01, v/v/v) at a flow rate of 1.0 mL/min. There was a good linearity over the range of 0.1-25 µg/mL (r = 0.9995) with a weighted (1/C(2) ) least square method. The lower limit of quantification was proved to be 0.1 µg/mL. The accuracy was within ±10.0% in terms of relative error and the intra- and inter-day precisions were less than 9.1% in terms of relative standard deviation. After validation, the method was successfully applied to characterize the pharmacokinetics of ponicidin in rats.
Osteoporosis is treated with oral and parenteral resorption inhibitors and parenteral osteogenic drugs. However, orally active small-molecule osteogenic drugs are not clinically available. Natural coumarin derivatives, such as osthole, exert osteoblastogenic effects.
A Sprague-Dawley rat model was utilized to elucidate perfluorooctanoic acid (PFOA), perfluorooctanesulfonic acid (PFOS) and perfluorohexanesulfonic acid (PFHxS) toxicokinetics with a goal of developing an in vivo approach for quantifying PFAS relative bioavailability in impacted soil. Following single dose administration (gavage) of ∼ 0.2-2000 µg kg BW of PFOA, PFOS or PFHxS, differences in PFAS blood, organ and excreta concentrations were observed over 120 h although linear dose responses were determined for area under the blood plasma time curves (AUC; PFOA, PFHxS), liver accumulation (LA: PFOS) and urinary excretion (UE; PFOA, PFHxS).
Platypuses are a unique freshwater mammal native to eastern Australia. They are semi-aquatic, predominantly nocturnal, and nest in burrows dug into the banks of waterbodies. Quantifying nesting burrow characteristics is challenging due to the species' cryptic nature.
The present study evaluated the antioxidant and anti-inflammatory properties of (CA) fruit extract against bisphenol A (BPA)-induced ovarian injury in female Wistar rats. The antioxidant activity was estimated by the total antioxidant capacity (TAC) and superoxide radical (NBT) content. For the in vivo analyses, 24 animals were divided into the following 4 groups: the control group; the BPA group (50 mg/kg BW BPA for 30 days); the BPA + CA group (50 mg/kg BW BPA and 50 mg/kg BW CA); and the CA group (50 mg/kg BW CA).
Background: Hemorrhagic shock (HS) corresponds to absolute hypovolemia creating an imbalance between oxygen supply and consumption. This causes an impaired hemostasis, a systemic inflammatory response, and microvascular permeability which can lead to multiple organ failure (MOF). There is no specific treatment for the endothelial dysfunction that plays a major role in the evolution towards MOF.
