The immunomodulatory effect of the antidepressant sertraline in an experimental autoimmune encephalomyelitis mouse model of multiple sclerosis.

Neuroimmunomodulation

Published: April 2011

Background: We have previously demonstrated the immunomodulatory activity of some selective serotonin reuptake inhibitors.

Objectives: In this research, we performed an in vivo/ex vivo study to evaluate the potential immunosuppressive effect of sertraline in an experimental autoimmune encephalomyelitis (EAE) model of multiple sclerosis.

Methods: Mice (C57/bl EAE) were treated with sertraline (5 mg/kg) or dexamethasone (1 mg/kg) 7 days after EAE induction and then 3 times weekly (for 3 weeks).

Results: Sertraline modestly improved the clinical score of EAE and attenuated the disease-induced weight loss. Sertraline also significantly decreased ex-vivo splenocyte viability, proliferation and secretion of pro-inflammatory cytokines in EAE mice.

Conclusions: We suggest that sertraline might be an add-on option for multiple sclerosis treatment.

Can fluoxetine influence orthodontic tooth movement? A systematic review and meta-analysis of studies on animal models.

Introduction: Selective serotonin reuptake inhibitors (SSRIs) such as fluoxetine adversely affect bone mineral density (BMD) and turnover, thereby increasing the risk of fractures. The objective of the present systematic review and meta-analysis was to evaluate studies on animal models that assessed whether fluoxetine can influence orthodontic tooth movement (OTM).

Article Synopsis
  • * Researchers tested sertraline, a common antidepressant, for its antifungal properties and found it reduced fungal levels in mice infected with C. auris, particularly in the kidneys.
  • * The combination of sertraline and voriconazole showed a stronger antifungal effect than either drug alone, suggesting a potential new treatment approach for C. auris infections.

Article Synopsis
  • The study investigates the role of the dorsolateral prefrontal cortex (DLPFC) in pain cognition among adolescents with depression, revealing abnormal cognitive responses to pain linked to this brain region.
  • Using neuroimaging data, the research compared functional connectivity (FC) of DLPFC in depressed adolescents to a control group and analyzed the effects of transcranial magnetic stimulation (TMS) versus sertraline treatment.
  • Results showed specific FC patterns correlated with pain perception and reduced pain-related thoughts after TMS treatment, suggesting that DLPFC is crucial in linking cognitive control and pain processing in affected adolescents.

In post-traumatic stress disorder (PTSD), anxiety-like symptoms are often associated with elevated noradrenaline levels and decreased serotonin. Selective serotonin reuptake inhibitors (SSRIs) are frequently used to treat anxiety, but elevated serotonin has been observed in some anxiety disorders. This study investigates stress-induced anxiety as an immediate effect of chronic stress exposure using the predator stress paradigm.

There are still few studies that have investigated the impact of sertraline (SE) on fertility, as well as adjuvant treatments that alleviate its side effects. Thus, the present study aimed to investigate the impact of SE on reproductive and neurobehavior parameters and verify whether the probiotic Lactobacillus rhamnosus alleviates the side effects of SE. After carrying out a dose-response study with SE, experiment II was conducted.
