Background: A novel approach for regulated bioanalytical sample preparation has been developed to combine multiple types of extraction techniques into one integrated and automated sample-preparation suite that pairs a graphical user interface with the Hamilton Microlab(®) STAR robotic liquid handler.
Results: The multi-assay sample-preparation suite is composed of three bioanalytical extraction techniques: protein precipitation, solid-phase extraction and liquid-liquid extraction. Validation data provided highly reproducible and robust results for each respective automated extraction technique.
Conclusion: The user-friendly graphical user interface and modular method design provide a flexible and versatile approach for routine bioanalytical sample-preparation and is the first fully integrated multiple assay sample-preparation suite for regulated bioanalysis.

Ambient mass spectrometry imaging (MSI) enables hundreds of analytes in tissue sections to be directly mapped at atmospheric pressure with minimal sample preparation. This field is currently experiencing rapid growth, with numerous reported ambient ionization techniques resulting in a "hundred flowers bloom" situation. Nanospray desorption electrospray ionization (nano-DESI), developed by the Laskin group in 2010, is a widely used liquid-extraction-based ambient ionization technique that was first used for mass spectrometry imaging of tissue in 2012.

Resonance Light Scattering (RLS) is a sensitive analytical technology hindered by its susceptibility to impurities in complex samples. This study introduces a combination of RLS with a high-efficiency sample preparation device, the Miniaturized Thermal-Assisted Purge-and-Trap (MTAPT), enhancing RLS's effectiveness in complex sample analysis. Specifically, we utilized MTAPT-RLS for the indirect screening of illegal hydrochloride drug additions in health products, based on three considerations: the transformation of bound HCl in hydrochloride drugs into volatile HCl under strong acid and heat; the minimal Cl content in health products for taste purposes; and the detectability of Cl ions by RLS upon the addition of AgNO and a stabilizer.
