A rapid and easy method to discriminate plant cultivars is indispensable to confirm food labeling. We established a fluorogenic ribonuclease protection (FRIP) assay to discriminate Japanese rice (Oryza sativa L.) cultivars based on single nucleotide polymorphisms (SNPs). The FRIP assay uses a hybridization technique between fluorescent probes and the target sequence prepared by run-off transcription, but the requirement of two PCR thermocycles is the problem when preparing template DNA for run-off transcription from rice genomic DNA. In this study, we designed new PCR primers with asymmetric melting temperatures. These primers amplified the target SNP marker containing a T7 RNA polymerase promoter sequence upstream of the target sequence in a single PCR. Moreover, 100 cultivars were discriminated with the patterns of 15 SNPs. The assay can be used as a rapid method of analysis to discriminate Japanese rice cultivars.

In terms of stress resistance, it has been suggested that plants gain or reinforce disease resistance during the process of maturation, which is recognized as adult plant resistance or age-related resistance. Polymer-coated fertilizers, widely used in rice cultivation in Japan, contribute to reactive nitrogen management and agricultural productivity but are a source of microplastics in the environment. Here, we investigated microplastics derived from polymer-coated fertilizer (microcapsule) runoff in Japanese paddy fields at 38 sites to quantitatively assess the behavior of microcapsules in paddy fields, and to estimate the total amount of runoff and accumulation in Japan. We also examined the factors causing variations in the amount of runoff among paddy fields.

Daki warm treatment (daki-ire) is performed during the process of seed mash preparation in the brewing of Japanese sake in order to promote the saccharification of rice by koji enzyme and to enhance the growth of Saccharomyces cerevisiae. Although it is important to control the growth of lactic acid bacteria in the preparation of kimoto-style seed mash (traditional sake-brewing method), it has not been known whether the transient increase in the temperature and/or appropriate temperature zone produced by daki-ire influences the growth of bacteria. A temperature increase generally helps bacterial growth, but we have found no published investigation of the influence of temperature changes in daki-ire on bacterial growth during the kimoto-style seed mash preparation process.
