The Milliflex Rapid System is a proven automated ATP-based solution used for rapid bioburden and sterility testing. Combining membrane filtration, ATP bioluminescence and imaging, it provides microbial enumeration in significantly less time than traditional methods. Detecting microbial contamination in cell-based products remains a challenge due to filterability issues and interference from mammalian ATP.

Metabolically active cells emit volatile organic compounds (VOCs) that can be used in real time to non-invasively monitor the health of cell cultures. We utilized these naturally occurring VOCs in an adapted culture method to detect differences in culturing Chinese hamster ovary (CHO) cells with and without Staphylococcus epidermidis and Aspergillus fumigatus contaminations. The VOC emissions from the cell cultures were extracted and measured from the culture flask headspace using polydimethylsiloxane (PDMS)-coated Twisters, which were subjected to thermal desorption-gas chromatography-mass spectrometry (TD-GC-MS) analysis.

Bulk electrical impedance spectroscopy (bio-capacitance) probes, hold significant promise for real-time cell monitoring in bioprocesses. Focusing on Chinese hamster ovary (CHO) cells, we present a sensitivity analysis framework to assess the impact of cell and culture properties on the complex permittivity spectrum, ε, and its associated parameters, permittivity increment, Δε, critical frequency, f, and Cole-Cole parameter, α, measured by bio-capacitance probes. Our sensitivity analysis showed that Δε is highly sensitive to cell size and concentration, making it suitable for estimating biovolume during the exponential growth phase, whereas f provides information about cumulative changes in cell size, membrane permittivity, and cytoplasm conductivity during the transition to death phase.

Methods: Using transcriptome sequencing, we analyzed gene expression in striped hamsters after short-term (3 days) and long-term (28 days) flea () parasitism.
