Introduction: Cerebral edema after ischemic stroke is frequently treated with mannitol and hypertonic saline (HS); however, their relative cerebrovascular and metabolic effects are incompletely understood, and may operate independent of their ability to lower intracranial pressure.

Methods: We compared the effects of 20% mannitol and 23.4% saline on cerebral blood flow (CBF), blood volume (CBV), oxygen extraction fraction (OEF), and oxygen metabolism (CMRO(2)), in nine ischemic stroke patients who deteriorated and had >2 mm midline shift on imaging. (15)O-PET was performed before and 1 h after administration of randomly assigned equi-osmolar doses of mannitol (1.0 g/kg) or 23.4% saline (0.686 mL/kg).

Results: Baseline CBF values (ml/100g/min) in the infarct core, periinfarct region, remaining ipsilateral hemisphere, and contralateral hemisphere in the mannitol group were 5.0 ± 3.9, 25.6 ± 4.4, 35.6 ± 8.6, and 45.5 ± 2.2, respectively, and in the HS group were 8.3 ± 9.8, 35.3 ± 10.9, 38.2 ± 15.1, and 35.2 ± 12.4, respectively. There was a trend for CBF to rise in the contralateral hemisphere after mannitol from 45.5 ± 12.2 to 57.6 ± 21.7, P = 0.098, but not HS. CBV, OEF, and CMRO(2) did not change after administration of either agent. Change in CBF in the contralateral hemisphere after osmotic therapy was strongly correlated with baseline blood pressure (R (2)= 0.879, P = 0.002).

Conclusions: We conclude that at higher perfusion pressures, osmotic agents may raise CBF in non-ischemic tissue. We conclude that at higher perfusion pressures, osmotic agents may raise CBF in non-ischemic tissue.

Objectives: Histological osteochondral characteristics of inflammation, fibrosis, vascularity, cartilage islands, vessels entering cartilage, thickened trabeculae and cysts are associated with bone marrow lesions (BMLs) in human knee osteoarthritis (OA). We identified and developed a method for scoring comparable pathology in two rat OA knee pain models.

Methods: Rats (n = 8-10 per group) were injected with monoiodoacetate (MIA) or saline, or underwent meniscal transection (MNX) or sham surgery.

Article Synopsis
  • This study investigates the use of esketamine, an NMDA receptor antagonist, to improve early recovery after shoulder arthroscopic surgery for rotator cuff injuries, focusing on pain and neurological dysfunction post-surgery.
  • The research involves 234 patients randomly assigned to receive varying doses of esketamine or a saline solution during general anesthesia, with early recovery quality assessed using the QoR-15 scale on the first and third postoperative days.
  • The trial aims to understand the drug’s effectiveness in reducing opioid use and postoperative complications like delirium and pain, while also examining its effects on intraoperative conditions such as cerebral oxygen saturation.

Purpose: The effects of coronavirus disease 2019 vaccination on fertility and pregnancy have turned out to be a main topic of public attention. Inactivated or recombinant protein vaccines are a reliable and safe method but mostly suffer from weak immunogenicity just in case formulated with a suitable adjuvant. The purpose of this research was to assess the impacts of new SpikoGen subunit vaccine administration during pregnancy on organogenesis in the rat fetus, which is a novel achievement in teratogenesis studies.

Objectives: Alcohol withdrawal syndrome manifests through a range of symptoms, including anxiety and anhedonia, significantly affecting the quality of life of those affected. This study investigates the potential therapeutic effects of the methanolic extract of leaves (MPG) on anxiety and anhedonia in Swiss albino female mice undergoing alcohol withdrawal.

Methods: Four groups of mice underwent alcohol withdrawal, with one group undergoing saline withdrawal as a control.

Enhancing protein O-GlcNAcylation by pharmacological inhibition of the enzyme O-GlcNAcase (OGA) is explored as a strategy to decrease tau and amyloid-beta phosphorylation, aggregation, and pathology in Alzheimer's disease (AD). There is still more to be learned about the impact of enhancing global protein O-GlcNAcylation, which is important for understanding the mechanistic path of using OGA inhibition to treat AD. In this study, we investigated the acute effect of pharmacologically increasing O-GlcNAc levels, using OGA inhibitor Thiamet G (TG), on normal mouse brains.
