Objective: To study the effect of mycophenolic acid (MPA) on maturation, endocytotic capacity and allostimulatory properties of human myeloid dendritic cell (MDC).
Methods: The peripheral blood mononuclear cells were isolated freshly from healthy volunteers (n = 15). The study group was treated by MPA. Co-stimulatory and adhesion molecular on MDC were analyzed by flow cytometry. After isolation of blood dendritic cell antigen-1(+) (BDCA-1(+)), the cellular uptake of FITC-dextran was measured by flow cytometry. After mixed lymphocyte reaction, the percentage of allogenic CD(4)(+) T cell in G(0) phase was analyzed by flow cytometry.
Results: (1) Maturation: Compared to control group, MPA down-regulated the level of CD40, CD62L, HLA-DR, CD54, CD80, CD83 and CD86 on MDC significantly in study group (t = -3.713, P > 0.010). (2) Endocytotic capacity: MPA enhanced significantly the endocytotic capacity of MDC in study group (t = 10.171, P = 0.000). (3) Allostimulatory capacity: Compared to control group, MPA reduced the ability of MDC to stimulate markedly the T cell proliferation in study group.
Conclusion: MPA may enhance the phagocytic capacity of human peripheral MDC, inhibit the maturity of MDC and impair its allostimulatory capacity of CD(4)(+) T lymphocyte.

Nanoplastics, solid polymer particles smaller than 1 μm, are suspected to be widely present in the environment, food and air, and may pose a potential threat to human health. Detecting nanoplastics in and associated with individual cells is crucial to understand their mechanisms of toxicity and potential harm. In this context, we developed a single-cell inductively coupled plasma time-of-flight mass spectrometry (sc-ICP-TOFMS) method for the sensitive and rapid quantification of metal-doped model nanoplastics in human cells.

Cargocomplexes play a vital role in non-viral delivery methods due to their capacity to target certain cells (or cells through the cell-division cycle) and inject their (macro)molecular "cargo" into them. The development of gene carriers that can efficiently transport and deliver genetic material into human-targeted cells with minimal toxicity is an important challenge in the field. The present study reports the straightforward preparation and testing of a modular non-viral gene carrier based on AuNPs.

Background: Immature dendritic cells (imDCs) play an important role in the induction of donor-specific transplant immunotolerance. However, these cells have limitations, such as rapid maturation and a short lifespan . In previous studies, induced pluripotent stem cells (iPSCs) differentiated into imDCs, and sinomenine (SN) was used to inhibit the maturation of imDCs.

Cyclosporin A nanocrystals (CsA-NCs) interaction with Caco-2 cells were investigated in this study, including cellular uptake and transport across Caco-2 cell monolayers. CsA-NCs of 165 nm, 240 nm and 450 nm were formulated. The dissolution of CsA-NCs was investigated by paddle method.

The O-GlcNAcylation is a posttranslational modification of proteins regulated by O-GlcNAc transferase (OGT) and O-GlcNAcase. These enzymes regulate the development, proliferation and function of cells, including the immune cells. Herein, we focused on the role of O-GlcNAcylation in human monocyte derived dendritic cells (moDCs).
