: Chronic liver diseases such as hepatic tumors can affect the brain through the liver-brain axis, leading to neurotransmitter dysregulation and behavioral changes. Cancer patients suffer from fatigue, which can be associated with sleep disturbances. Sleep is regulated via two interlocked mechanisms: homeostatic regulation and the circadian system.

Article Synopsis
  • Adar2 mice are commonly used to study the effects of reduced RNA editing, specifically relating to the Gria2 subunit of the AMPA receptor, which impacts their circadian rhythms.
  • The study found that Gria2 mice lost circadian rhythmicity in the hippocampus when compared to Adar2 mice, indicating a significant effect on their internal clock.
  • These findings suggest that the postnatal increase in editing of the Gria2 subunit is important for the circadian clock's development in certain brain areas, raising questions about using Gria2 mice as controls in related experiments.

The circadian clock is one of the most important homeostatic systems regulating the majority of physiological functions. Its proper development contributes significantly to the maintenance of health in adulthood. Methadone is recommended for the treatment of opioid use disorders during pregnancy, increasing the number of children prenatally exposed to long-acting opioids.

Nicotinamide adenine dinucleotide (NAD) is an essential cofactor of critical enzymes including protein deacetylase sirtuins/SIRTs and its levels in mammalian cells rely on the nicotinamide phosphoribosyltransferase (NAMPT)-mediated salvage pathway. Intracellular NAMPT (iNAMPT) is secreted and found in the blood as extracellular NAMPT (eNAMPT). In the liver, the iNAMPT-NAD axis oscillates in a circadian manner and regulates the cellular clockwork.

Chronic liver diseases including hepatocellular carcinoma (HCC) create a state of chronic inflammation that affects the brain via the liver-brain axis leading to an alteration of neurotransmission and cognition. However, little is known about the effects of HCC on the hippocampus, the key brain region for learning and memory. Moreover, radiotherapy used to treat HCC has severe side effects that impair patients' life quality.
