We report on the application of an optically tunable coherent white light source, based on the supercontinuum generation from microstructured optical fibres, to fluorescence lifetime imaging (FLIM) microscopy and Foerster resonance energy transfer (FRET). A prototype lens based on monotonic longitudinal chromatic aberration is used for tuning the supercontinuum wavelengths from 400 to 1000 nm and acts as an axial monochromator, suitable for fibre delivery in confocal microscopy.

1Parkinson's disease (PD) involves the aggregation of the protein alpha-synuclein, a process promoted by interactions with intracellular membranes. To study this phenomenon in neurons for the first time, we developed a fluorescence lifetime imaging (FLIM) method using Förster resonance energy transfer and self-quenching reporters, analyzed with a custom-built FLIM microscope. This method offers insights into aggregate formation in PD and can be broadly applied to probe protein-membrane interactions in neurons.

Purpose: Current technologies to define the zone of acute peripheral nerve injury intraoperatively are limited by surgical experience, time, cumbersome electrodiagnostic equipment, and interpreter reliability. In this pilot study, we evaluated a real-time, label-free optical technique for intraoperative nerve injury imaging. We hypothesize that fluorescence lifetime imaging (FLIm) will detect a difference between the time-resolved fluorescence signatures for acute crush injuries versus uninjured segments of peripheral nerves in sheep.
