Based on fourth generation diaminobutane poly(propylene imine) dendrimer, a novel targeted drug nanocarrier was prepared, bearing protective PEG chains and a folate targeting ligand. As a control a PEGylated derivative without folate was also synthesized. The encapsulation and release properties of these PEGylated derivatives were investigated employing etoposide, an anticancer hydrophobic drug. Enhanced solubility of etoposide was achieved inside the dendrimeric scaffold which was subsequently released in a controlled manner. These properties coupled with specificity towards the folate receptor and the low toxicity render folate functionalized PEGylated poly(propylene imine) dendrimer promising candidate for targeted drug delivery.

Controlled pore glass (CPG), differing in pore size and subsequent specific surface, was chemically modified by: (1) increasing surface susceptibility for amine functionalization via reaction with oxirane-type (active) and alkyl/aryl-type (inactive towards amine compounds) silane pro-adhesive compounds, and (2) immobilization of trimethylolpropane tris[poly(propylene glycol), amine terminated] ether, comb-like 8-arm octa[poly(ethylene glycol) amine] with each branch amine terminated, and a poly(propylene imine) amine-terminated second-generation dendrimer. The increase in surface density of amine functions - monitored by UV-Vis technique adopted for quantitative measurements of Ruhemann's purple intensity - improved final loading capacity, characterized by dimethoxytrityl cation absorption. Obtained materials proved their applicability in automatic oligonucleotide (ON) synthesis, especially when silanized 2000 Å CPG modified with 8-arm octa[poly(ethylene glycol) amine], with deduced empirical formula CPG - silane - (NH)PEG-(NH), was used for long-chain (150 nucleotides) ONs synthesis.

Polymer microstructures rely on tacticity, yet exploration in polyamines has focused predominantly on atactic polymers. We introduce a method to synthesize a diverse library of and -cyanobenzenesulfonyl-activated-methyl aziridines using , , and racemic alaninol. Living anionic ring-opening polymerization of racemic sulfonyl aziridines yields soluble polymers, while enantiomerically-pure sulfonyl aziridines follow a dispersion polymerization with complete monomer conversion giving access to stereoblock copolymers.

Glomerulus-Targeted ROS-Responsive Polymeric Nanoparticles for Effective Membranous Nephropathy Therapy.

Membranous nephropathy (MN) is a common immune-mediated glomerular disease that requires the development of safe and highly effective therapies. Celastrol (CLT) has shown promise as a therapeutic molecule candidate, but its clinical use is currently limited due to off-target toxicity. Given that excess levels of reactive oxygen species (ROS) contributing to podocyte damage is a key driver of MN progression to end-stage renal disease, we rationally designed ROS-responsive cationic polymeric nanoparticles (PPS-CPNs) with a well-defined particle size and surface charge by employing poly(propylene sulfide)-polyethylene glycol (PPS-PEG) and poly(propylene sulfide)-polyethylenimine (PPS-PEI) to selectively deliver CLT to the damaged glomerulus for MN therapy.

Spray-drying of PEI-/PPI-based nanoparticles for DNA or siRNA delivery.

Spray-drying of nucleic acid-based drugs designed for gene therapy or gene knockdown is associated with many advantages including storage stability and handling as well as the possibility of pulmonary application. The encapsulation of nucleic acids in nanoparticles prior to spray-drying is one strategy for obtaining efficient formulations. This, however, strongly relies on the definition of optimal nanoparticles, excipients and spray-drying conditions.

Implications of Defect Density and Polymer Interactions for CO Capture on Amine-Functionalized MIL-101(Cr).

Department of Chemical and Biological Engineering, Princeton University, 41 Olden Street, Princeton, New Jersey, 08544, USA.

Rising anthropogenic carbon emissions have dire environmental consequences, necessitating remediative approaches, which includes use of solid sorbents. Here, aminopolymers (poly(ethylene imine) (PEI) and poly(propylene imine) (PPI)) are supported within solid mesoporous MIL-101(Cr) to examine effects of support defect density on aminopolymer-MOF interactions for CO uptake and stability during uptake-regeneration cycles. Using simulated flue gas (10 % CO in He), MIL-101(Cr)-ρ (higher defect density) shows 33 % higher uptake capacity per gram adsorbent than MIL-101(Cr)-ρ (lower defect density) at 308 K, consistent with increased availability of undercoordinated Cr adsorption sites at missing linker defects.
