Article Synopsis
  • At week 48 of the DAWNING study, dolutegravir plus two nucleoside reverse transcriptase inhibitors showed better virologic suppression than ritonavir-boosted lopinavir in adults with HIV-1 who had previously failed treatment.
  • Among the 314 participants, 7 developed treatment-emergent INSTI resistance-associated mutations, indicating some resistance to dolutegravir, though they still achieved low HIV-1 RNA levels.
  • The study reveals that while mutations can confer some resistance to dolutegravir, this resistance comes with a significant trade-off: a decrease in the virus’s ability to replicate effectively.

: The oligomerization of HIV-1 integrase onto DNA is not well understood. Here we show that HIV-1 integrase binds the DNA in biphasic (high-affinity and low-affinity) modes. For HIV-1 subtype B, the high-affinity mode is ∼100-fold greater than the low-affinity mode (Kd.

Retroviral intasomes arising.

Curr Opin Struct Biol

Retroviral DNA integration takes place in the context of the intasome nucleoprotein complex. X-ray crystal structures of functional spumaviral intasomes were previously revealed to harbor a homotetramer of integrase, and it was generally believed that integrase tetramers catalyzed the integration of other retroviruses. The elucidation of new structures from four different retroviruses over the past year has however revealed this is not the case.

Crystal structure of the Rous sarcoma virus intasome.

Nature

Department of Biochemistry, Molecular Biology and Biophysics, University of Minnesota, Minneapolis, Minnesota 55455, USA.

Article Synopsis
  • The integration of reverse-transcribed viral DNA into a host genome is crucial for retroviruses, with integrase playing a key role in this process.
  • Structures of integrase-DNA complexes have been studied, but details of a three-domain integrase bound to DNA were previously unclear.
  • This study reveals the crystal structure of Rous sarcoma virus integrase, showing an octameric assembly that facilitates its complex interaction with viral and target DNAs, highlighting the varied roles of its domains and advancing our understanding of retrovirus integration mechanisms.

Due to the importance of human immunodeficiency virus type 1 (HIV-1) integrase as a drug target, the biochemistry and structural aspects of retroviral DNA integration have been the focus of intensive research during the past three decades. The retroviral integrase enzyme acts on the linear double-stranded viral DNA product of reverse transcription. Integrase cleaves specific phosphodiester bonds near the viral DNA ends during the 3' processing reaction.
