Isocratic methods are described for the separation of purine compounds in specimens of jejunal mucosa available by peroral biopsy from patients with coeliac disease. The first mode provided separation of both the ionic nucleoside monophosphates and the non-ionic nitrogen bases and nucleosides. The second mode was applicable to analyses of the nucleoside mono-, di- and triphosphates only. These chromatographic procedures were carried out with tetrabutylammonium phosphate as an ion-pair modifier and different concentrations of acetonitrile in the mobile phase. The modes are flexible in that they permit the optimization of the separation of these compounds according to the purpose of the investigation. The results of studies on purine metabolite patterns in coeliac mucosa are discussed.

This study introduces a novel synchronous spectrofluorimetry coupled with chemometric tools for the determination of tenofovir and dolutegravir antiretroviral drugs. Utilizing partial least squares regression (PLS) fine-tuned by genetic algorithm as variable selection tool, the developed models demonstrate greater sensitivity, cost-effectiveness, and reduced environmental impact compared to traditional HPLC methods. The model's validation was further confirmed using external validation in addition to QC samples as per ICH M10 guidelines, which yielded high accuracy ranged between 94.

In this study, we analyzed purine derivatives using multimatrix variation matrix-assisted laser desorption ionization mass spectrometry (MALDI MS) with α-cyano-4-hydroxycinnamic acid (CHCA), 1,5-diaminonaphtalene (DAN), 5-formylsalicylic acid (FSA), and 5-nitrosalicylic acid (NSA) as matrices. Further, we focused on the abstraction/attachment of hydrogen from/to analytes and detected [M - H], [M + 2H] and/or [M + 3H] in MALDI MS spectra of compounds containing nitrogen and/or carbonyl oxygen. Although [M - H] generation of purine compounds in MALDI MS with conventional matrices was challenging, NSA-MALDI MS effectively yielded the [M - H]species of purine derivatives compared with CHCA, FSA, and DAN, and the [M - H]/[M + H] ratios reflected their structures, such as the substituting groups and positions.
