The binding modes of noncompetitive GABA(A)-channel blockers were re-examined taking into account the recent description of the 3D structure of prokaryotic pentameric ligand-gated ion channels, which provided access to new mammalian or insect GABA receptor models, emphasizing their transmembrane portion. Two putative binding modes were deciphered for this class of compounds, including the insecticide fipronil, located nearby either the intra- or the extracellular part of the membrane, respectively. These results are in full agreement with previously described affinity-labeling reactions performed with GABA(A) noncompetitive blockers (Perret et al. J. Biol. Chem.1999, 274, 25350-25354).

This research investigates the thin film formation of the Cu-BDC (copper benzene-1,4-dicarboxylate) MOF system on a carboxylic acid-terminated self-assembled monolayer by alternating deposition of solution-phase inorganic and organic precursors. Nonequilibrium membrane pattern formation is studied using meshless membrane simulation. We consider that molecules bind to either surface of a bilayer membrane and move to the opposite leaflet by flip-flop. When binding does not modify the membrane properties and the transfer rates among the three states are cyclically symmetric, the membrane exhibits spiral-wave and homogeneous-cycling modes at high and low binding rates, respectively, as in an off-lattice cyclic Potts model.

Transcription activators are said to stimulate gene expression by 'recruiting' coactivators, yet this vague term fits multiple kinetic models. To directly analyze the dynamics of activator-coactivator interactions, single-molecule microscopy was used to image promoter DNA, a transcription activator and the Spt-Ada-Gcn5 acetyltransferase (SAGA) complex within yeast nuclear extract. SAGA readily but transiently binds nucleosome-free DNA without an activator, while chromatin association occurs primarily when an activator is present.
