Elongation factor 4 is a widely distributed translational GTPase also known as LepA. Its physiological role is ambiguous, as only a few phenotypes resulting from lepA null mutations have been reported. Here, we report that a Streptomyces coelicolor lepA null mutant overproduces the calcium-dependent antibiotic (CDA). Our findings are the first that connect LepA (encoded by SCO2562) to antibiotic production. They lend additional evidence that perturbations in the quaternary structure and function of the ribosome can positively affect antibiotic production in Streptomyces bacteria.

In Australia, pyrethrum (Tanacetum cinerariifolium) cultivation provides a significant portion of the global supply of natural insecticidal pyrethrins. However, crown and root rots, along with stunted plant growth and plant loss during winter, are significant issues affecting certain sites. Several isolates of the Fusarium oxysporum species complex (FOSC) have been identified as causal agents of crown and root rot in pyrethrum, highlighting these as key pathogens contributing to this decline.

In this study, we revealed a critical role of eukaryotic elongation factor-2 kinase (eEF-2K), a negative regulator of protein synthesis, in regulating T cells during vaccinia virus (VACV) infection. We found that eEF-2K-deficient (eEF-2K⁻/⁻) mice exhibited a significantly higher proportion of VACV-specific effector CD8 T cells without compromising the development of VACV-specific memory CD8 T cells. RNA sequencing demonstrated that eEF-2K⁻/⁻ VACV-specific effector CD8 T cells had enhanced functionality, which improves their capacity to combat viral infection during the effector phase.
