ROR-alpha is an orphan nuclear receptor, inactivation of which cell-autonomously blocks differentiation of cerebellar Purkinje cells with a secondary loss of granule neurons. As part of our ENU mutagenesis screen we isolated the recessive tmgc26 mouse mutant, characterized by early-onset progressive ataxia, cerebellar degeneration and juvenile lethality. Detailed analysis of the tmgc26-/- cerebella revealed Purkinje cell and granule cell abnormalities, and defects in molecular layer interneurons and radial glia. Chimera studies suggested a cell-autonomous effect of the tmgc26 mutation in Purkinje cells and molecular layer interneurons, and a non-cell-autonomous effect in granule cells. The mutation was mapped to a 13-Mb interval on chromosome 9, a region that contains the ROR-alpha gene. Sequencing of genomic DNA revealed a T-to-A transition in exon 5 of the ROR-alpha gene, resulting in a nonsense mutation C257X and severe truncation of the ROR-alpha protein. Together, our data identify new roles for ROR-alpha in molecular layer interneurons and radial glia development and suggest tmgc26 as a novel ROR-alpha allele that may be used to further delineate the molecular mechanisms of ROR-alpha action.

BACKGROUND Cowden syndrome is a genetic disorder that predisposes individuals to cancer and is characterized by hamartomas derived from 3 germ layers. Although the clinical signs can be pathognomonic, diagnosis is often aided by biopsies, histopathological examination of oral and cutaneous lesions, and genetic studies, including multiple ligation-dependent probe amplification (MLPA). CASE REPORT We report a case of a 35-year-old woman who manifested with multiple lesions in the buccal mucosa, dorsum of the tongue, and gums, along with papillomatous papules on her facial skin and the dorsal surfaces of her hands.

Here we describe the synthesis and evaluation of a molecular corrosion sensor that can be applied in situ in aerospace coatings, then used to detect corrosion after the coating has been applied. A pH-sensitive molecule, 4-mercaptopyridin (4-MP), is attached to a gold nanoparticle to allow surface-enhanced Raman-scattering (SERS) for signal amplification. These SERS nanoparticles, when combined with an appropriate micron-sized carrier system, are incorporated directly into an MIL-SPEC coating and used to monitor the process onset and progression of corrosion using pH changes occurring at the metal-coating interface.

In this study, it is shown that an efficient organic optocoupler (OPC) can be fabricated using commercially available and solution-processable organic semiconductors. The transmitter is a single-active-layer organic light-emitting diode (OLED) made from a well-known polyparavinylene derivative, Super Yellow. The receiver is an organic light-emitting diode (OLSD) with a single active layer consisting of a mixture of the polymer donor PTB7-Th and the low-molecular-weight acceptor ITIC; the receiver operates without an applied reverse voltage.

The surfaces of beech wood samples were treated with polyethylenimine (PEI) solutions at three different concentrations-0.5%, 1% and 2%-and two molecular weights-low molecular weight (LMW) and high molecular weight (HMW). Using the framework of an investigation of the stimuli-responsive behavior of peptide assembly on a solid surface, this study on the behavior of a chemisorbed peptide on a gold surface was performed. The studied peptide is a dimeric form of the antimicrobial peptide Trichogin GAIV, which was also modified by substituting the glycine with lysine residues, while the N-terminus octanoyl group was replaced by a lipoic one that was able to bind to the gold surface. In this way, a chemically linked peptide assembly that is pH-responsive was obtained because of the protonation/deprotonation of the sidechains of the Lys residues.
