The authors have studied the value of culture media based on pyruvic acid (the Dixon medium and an original medium called P.T.) in the isolation of mycobacteria from sputum, as compared with the Löwenstein-Jensen medium. The studies were carried out in two groups of patients: 1043 samples were collected from ambulatory patients and 1740 samples were collected from hospitalized patients. The superiority of the Dixon and of the P.T. media was confirmed by the increased percentage of positive cultures (11% and 20% respectively) as compared with the Löwenstein-Jensen medium. With these media it was possible to isolate mycobacteria 7--14 days earlier than with the other media. On the basis of the results obtained the authors recommend the use, in parallel, especially in those cases when the samples contain only a small number of bacteria, of the Löwenstein-Jensen medium and one of the media based on pyruvic acid.

Background: Osteopenia (ON) and osteoporosis (OP) are highly prevalent among postmenopausal women and poses a challenge for early diagnosis. Therefore, identifying reliable biomarkers for early prediction using metabolomics is critically important.
Methods: Initially, non-targeted metabolomics was employed to identify differential metabolites in plasma samples from cohort 1, which included healthy controls (HC, n = 23), osteonecrosis (ON, n = 36), and osteoporosis (OP, n = 37).
Head and neck squamous cell carcinoma (HNSCC) develops and advances because of the accumulation of somatic mutations located in orthosteric and allosteric areas. However, the biological effects of allosteric driver mutations during tumorigenesis are mostly unknown. Here, we mapped somatic mutations generated from 10 tumor-normal matched HNSCC samples into allosteric sites to prioritize the mutated allosteric proteins via whole-exome sequencing and AlloDriver, identifying the specific mutation H351Q in β-glucuronidase (GUSB), a lysosomal enzyme, as a novel allosteric driver mutation, which considerably encouraged HNSCC progression both in vitro and in vivo.
