We report that dengue virus (DENV) methyltransferase sequentially methylates the guanine N-7 and ribose 2'-O positions of viral RNA cap (GpppA-->m(7)GpppA-->m(7)GpppAm). The order of two methylations is determined by the preference of 2'-O methylation for substrate m(7)GpppA-RNA to GpppA-RNA, and the 2'-O methylation is not absolutely dependent on the prior N-7 methylation. A mutation that completely abolished the 2'-O methylation attenuated DENV replication in cell culture, whereas another mutation that abolished both methylations was lethal for viral replication, suggesting that N-7 methylation is more important than 2'-O methylation in viral replication. The latter mutant with lethal replication could be rescued by trans complementation using a wild-type DENV replicon. Furthermore, we found that chimeric DENVs containing the West Nile virus methyltransferase, polymerase, or full-length NS5 were nonreplicative, but the replication defect could also be rescued through trans complementation using the wild-type DENV replicon.

This work describes the synthesis of ordered 3D siloxane-silsesquioxane reticular materials with silicate D4R cubes (SiO), harvested from a sacrificial tetrabutylammonium cyclosilicate hydrate (TBA-CySH) precursor, interlinked with octyl and dicyclopentyl (Cp) hydrocarbon functionalities in a one-step synthesis with organodichlorosilanes. Advanced solid-state NMR spectroscopy allowed us to unravel the molecular order of the nodes and their interconnection by the silicone linkers. In the case of octyl-methyl silicone linkers, changing the silane-to-silicate ratio in the synthesis allowed for tuning the length of the linker between the nodes.

Due to the endocrine toxicity, neurotoxic, and reproductive toxicity to organisms, the sources and risks of brominated organic pollutants have attracted widespread attention. However, knowledge gaps remain in the bromination processes of emerging phenolic pollutants in plants, which may increase the potential health risk associated with food exposure. Our study discovered that light induced generation and accumulation of more toxic brominated organic compounds (Br-org) in lettuce leaves under the stress of acetaminophen (ACE) than that without light, as evidenced by an increase in C-Br bond intensity in FTIR analysis.

We developed a unique water droplet templating method to fabricate polymer films with three-dimensionally ordered porous structures. This technique is based on a polymer/solvent/HO ternary system, and the key is to choose a volatile and hydrophobic solvent that is slightly miscible with HO. With the fast evaporation of the solvent, water droplets separate from the casting solution and condense from the air to act as pore templates inside the film and on the surface, respectively.
