It is well known that the seeds of watermelon [Citrullus lanatus (Thunb.) Matsum and Nakai] have a high potential to germinate when the fruit has ripened. When removed from the mature fruit, the seeds can germinate under appropriate conditions. However, it is unclear why they cannot germinate in the flesh of the fruit. Here, we show that cis-ABA and its β-D-glucopyranosyl ester (ABA-β-GE) accumulate in the flesh of the fruit at levels high enough to inhibit seed germination. This result indicates the existence of chemical factors that inhibit viviparous seed germination of watermelon.

The firmness of the two apple varieties: Idared and Pinova was similar during ripening, while it decreased significantly during 3-month storage only for Idared. Pectin-rich fractions were isolated from apple flesh tissue: water-soluble pectin (WSP), imidazole-soluble pectin (ISP), and hemicellulose-rich fractions: natively acetylated hemicelluloses (LiCl-DMSO), deacetylated hemicelluloses (KOH). It was shown that the degree of acetylation (DAc) of the hemicelluloses fraction (LiCl-DMSO) increased during apple ripening and storage, with higher values for Idared.

To clarify the cause of graded distribution of sucrose in apple fruit flesh, a quarter cut of young apple fruit was cultured for 72 h on agar-solidified MS medium supplemented with 0.5 M [1-C]sorbitol, with the longitudinal or horizontal cut face being attached with the medium, and distribution of C-labelled sucrose in a specimen obtained by slicing the fruit along with the cut face was visualized utilizing MALDI-TOF MSI. Heat map images on the distribution of the peaks of sorbitol containing C-atom indicated that external [1-C]sorbitol had penetrated evenly into the tissue.
