Ultra-high-molecular-weight poly-gamma-glutamic acid (gamma-PGA) is a most promising biodegradable polymer that is produced by Bacillus subtilis (chungkookjang). Attractive properties of gamma-PGA are that it is water soluble, anionic, biodegradable, and edible. Development of gamma-PGA has pursued in terms of cosmetics/skin care, bone care, nanoparticle for drug delivery system, hydrogel, and so on. Very recently, our research has shown that gamma-PGA can be used as an immune-stimulating agent, especially at high molecular weight. This review presents the synthesis and production of high-molecular-weight gamma-PGA and its various applications in industrial fields.

Interferon-γ (IFN-γ) is a key mediator in antitumor immunity and immunotherapy responses, yet its clinical applications remain restricted to chronic granulomatous disease and malignant osteopetrosis. IFN-γ effectiveness as a standalone treatment has shown limited success in clinical trials and its potential for synergistic effects when combined with immunotherapies is under clinical exploration. A particularly compelling combination is that of IFN-γ with Toll-like receptor (TLR) agonists that holds significant promise for cancer treatment.

The presented work discusses the highly efficient esterification of poly (γ-glutamic acid) (γ-PGA) with alkyl halides at room temperature. The esterification reaction was completed within 3 h, and the prepared γ-PGA esters were obtained with excellent yields (98.6%) when 1,1,3,3-tetramethylguanidine (TMG) was used as a promoter.

Poly-γ-glutamic acid (γ-PGA) is a natural polymer whose molecular weight and viscosity are critical for its application in various fields. However, research on super-high-molecular-weight or -viscosity γ-PGA is limited. In this study, the gene in WX-02 was knocked out using homologous recombination, and the batch fermentation performances of the recombinant strain WX-ΔpgdS were compared to those of WX-02.

: To explore the relationship between the stability of poly(gamma-glutamic acid) (γ-PGA) dispersion systems with γ-PGA of different molecular weights (MWs) and concentrations and type I collagen mineralization. : γ-PGA was used as a noncollagenous protein (NCP) analogue to regulate the stability of supersaturated γ-PGA-stabilized amorphous calcium phosphate (PGA-ACP) solutions by changing the γ-PGA MW (2, 10, 100, 200 and 500 kDa) and concentration (400, 500 and 600 μg mL). Then, the optical density (OD) at 72 h was measured to determine the PGA-ACP solution stability.

Senescence and osteogenic differentiation potential loss limited bone nonunion treatment effects of bone marrow-derived mesenchymal stem cells (BMSCs). MiR-100-5p/Lysine(K)-specific demethylase 6B (KDM6B) can inhibit osteogenesis, but their effects on bone union remain unclear. This study aims to investigate the effects of miR-100-5p/KDM6B on osteogenic differentiation and bone defects.
