Functional somatic syndromes include some of the most common and frustrating illnesses seen by primary care physicians and medical specialists. An extensive literature search of the 2 best characterized functional somatic syndromes, fibromyalgia and irritable bowel syndrome, reveals the overlap of these 2 disorders and their close relationship to depression. New pathophysiologic studies have shown that there are similar central nervous system changes in fibromyalgia, irritable bowel syndrome, and depression. These clinical and biologic similarities are consistent with the observations that the effective management of fibromyalgia and irritable bowel syndrome is comparable to that of depression.

Background: The goal of this study was to examine autonomic nervous system function by measuring heart rate (HR), heart rate variability (HRV), skin conductance levels (SCL), and peripheral skin temperature (ST) in response to and during recovery from psychosocial stressors in patients with functional somatic syndromes (FSS; fibromyalgia and/or chronic fatigue syndrome), stress-related syndromes (SRS; overstrain or burn-out), and healthy controls (HC).

Methods: Patients with FSS (n = 26), patients with SRS (n = 59), and HC (n = 30) went through a standardized psychosocial stress test consisting of a resting phase (120 s), the STROOP color word task (120 s), a mental arithmetic task (120 s) and a stress talk (120 s), each followed by a 120 s recovery period. HR, HRV, SCL, and ST were monitored continuously.

ZIC1 is a context-dependent medulloblastoma driver in the rhombic lip.

Transcription factors are frequent cancer driver genes, exhibiting noted specificity based on the precise cell of origin. We demonstrate that ZIC1 exhibits loss-of-function (LOF) somatic events in group 4 (G4) medulloblastoma through recurrent point mutations, subchromosomal deletions and mono-allelic epigenetic repression (60% of G4 medulloblastoma). In contrast, highly similar SHH medulloblastoma exhibits distinct and diametrically opposed gain-of-function mutations and copy number gains (20% of SHH medulloblastoma).

Glioblastoma is a highly aggressive primary brain tumor with glioblastoma stem cells (GSCs) enforcing the intra-tumoral hierarchy. Plasma cells (PCs) are critical effectors of the B-lineage immune system, but their roles in glioblastoma remain largely unexplored. Here, we leverage single-cell RNA and B cell receptor sequencing of tumor-infiltrating B-lineage cells and reveal that PCs are aberrantly enriched in the glioblastoma-infiltrating B-lineage population, experience low level of somatic hypermutation, and are associated with poor prognosis.
