In this study, we checked whether HSP70 preparations of different origins are able to protect model animals (rats) from endotoxic shock and modify the response of myeloid cells to lipopolysaccharide (LPS) challenge. It was shown that HSP70 preparations can effectively protect organisms from endotoxic shock by strongly decreasing mortality and restoring both homeostasis and various hemodynamic characteristics. At the cellular level, HSP70 preparations significantly inhibit LPS-induced reactive oxygen species production in various myeloid cells and decrease NO expression in macrophages, which is enhanced after LPS priming. In parallel, HSP70 preconditioning partially normalizes neutrophil apoptosis, which is disturbed as a result of LPS stimulation. These results suggest that the antiseptic actions of HSP70 preparations are probably realized at the level of receptor membrane complexes of myeloid cells, which represent the major target of LPS action. Taken together, our findings show that extracellular mammalian HSP70 may play an important role in innate immunity modulation and stimulation of endogenous protective mechanisms, both at the cellular and organism levels, which make this protein a promising base for the development of efficient antiseptic drugs.

Photothermal therapy is a novel and promising method for cancer treatment due to its controllable property, noninvasive nature, and high selectivity. Nevertheless, tumor recurrence of inflammatory response and tumor tolerance of heat shock protein over-expression remain serious challenges in current photothermal therapy. Additionally, the high dosage requirement of nanomaterial for optimal imaging and therapeutic effect would result in various side effects, organ excretion burdens, and long-term accumulation in the body.

HSP70 is upregulated after heat but not freezing stress in the freeze-tolerant cricket Gryllus veletis.

Heat shock proteins (HSPs) are well known to prevent and repair protein damage caused by various abiotic stressors, but their role in low temperature and freezing stress is not well-characterized in insects compared to other thermal challenges such as heat stress. Ice formation in and around cells is hypothesized to cause protein damage, yet many species of insects can survive freezing, suggesting HSPs may be an important mechanism in freeze tolerance. Here, we studied HSP70 in a freeze-tolerant cricket Gryllus veletis to better understand the role of HSPs in this phenomenon.

Dietary seaweed extract mitigates oxidative stress in Nile tilapia by modulating inflammatory response and gut microbiota.

Nutrition and Seafood Laboratory (NuSea.Lab), School of Life and Environmental Sciences, Deakin University, Queenscliff, VIC, Australia.

Introduction: Extreme water temperature affects the well-being of all aquatic animals, including fish. Higher temperatures can lead to the generation of reactive oxygen species (ROS), which can induce oxidative stress and negatively impact fish health and well-being. This study investigated the protective effects of seaweed extract on growth, antioxidant status, inflammatory responses, and gut microbiota to gain a better understanding of the acclimatization ability of Nile tilapia, in response to oxidative stress caused by high water temperatures.

Human peripheral blood mononuclear cells (PBMCs) are used to examine biological processes and disease, when basal variability in cellular activation and splicing is described and unexplained. Using isolation systems that maintained buffy coat cells (PBMCs, platelets) in their own plasma, poly-A enriched RNA-sequencing (RNASeq) detected 42,720 Ensembl gene IDs, including >95% of the top 100 Genotype Tissue Expression Project (GTEx)-expressed genes in lung, colon, heart, skeletal muscle and liver, and 10/17 clinically-actionable genes listed by the Pharmacogenomics Knowledgebase. Transcriptome changes were defined after 1h treatment with 32°C hypothermia (hsp70 family member change), 10 μmol/L ferric citrate that had no discernible effect, and 100 μg/mL cycloheximide leading to induction of primary response (immediate early) genes including IL1B and TNF.

Article Synopsis
  • * Researchers tested various concentrations of ethanol and water extracts from leaves and stems for their ability to enhance cell viability and reduce oxidative stress markers.
  • * Results showed that specific concentrations of leaf and stem extracts effectively prevented cell death, improved mitochondrial function, and could have potential protective effects against neurodegenerative conditions.
