The elaboration of biologically important 3,4-substituted pyrazolines was achieved by an organocatalysed aza-Michael/transimination domino sequence between hydrazones and enones making use of a mixture of heterogeneous resin-bound acid/base reagents. This methodology nicely illustrates the site isolation concept of supported reagents allowing the simultaneous use of otherwise destructive reactive functionalities.

An isomerization reaction of 7-aryl-3-formylpyrazolo[1,5-]pyrimidines to 5-aroyl-NH-pyrazolo[3,4-]pyridines proceeding with high yields in aqueous NaOH under microwave conditions is reported. This unprecedented transformation occurs by adding and eliminating a water molecule an ANRORC mechanism (adding the nucleophile, ring-opening, and ring-closing) studied using DFT calculations. The product's utility was proved as they have aroyl and NH groups that simple methods and readily available reagents easily modified; likewise, their optical properties were studied, highlighting their high potential as highly emissive modular dyes ( up to 99%).

The fused heterocycle 1-(imidazo[5,1-a]isoquinolin-3-yl)naphthalen-2-ol (LH) has been synthesized and characterized by spectroscopic methods. Probe LH upon irradiation with λ = 336 nm exhibited strong fluorescence with λ = 437 nm in MeOH/HEPES buffer (5 mM, pH = 7.4, 2:8, v/v).

Chemical modifications are widely used in research fields such as quantitative proteomics and interaction analyses. Chemical-modification targets can be roughly divided into four categories, including those that integrate isotope labels for quantification purposes, probe the structures of proteins through covalent labeling or cross-linking, incorporate labels to improve the ionization or dissociation of characteristic peptides in complex mixtures, and affinity-enrich various poorly abundant protein translational modifications (PTMs). A chemical modification reaction needs to be simple and efficient for use in proteomics analysis, and should be performed without any complicated process for preparing the labeling reagent.
