Chemical aminoacylation of tRNAs with fluorinated amino acids for in vitro protein mutagenesis.

Published: April 2010

This article describes the chemical aminoacylation of the yeast phenylalanine suppressor tRNA with a series of amino acids bearing fluorinated side chains via the hybrid dinucleotide pdCpA and ligation to the corresponding truncated tRNA species. Aminoacyl-tRNAs can be used to synthesize biologically relevant proteins which contain fluorinated amino acids at specific sites by means of a cell-free translation system. Such engineered proteins are expected to contribute to our understanding of discrete fluorines' interaction with canonical amino acids in a native protein environment and to enable the design of fluorinated proteins with arbitrary desired properties.

STRUCTURAL AND FUNCTIONAL BONE FEATURES IN CHILDREN RESIDING IN THE RADIOLOGICALLY CONTAMINATED TERRITORIES OF UKRAINE.

Probl Radiac Med Radiobiol

December 2024

State Institution «National Research Center of Radiation Medicine, Hematology and Oncology of the National Academy of Medical Sciences of Ukraine», 53 Yuriia Illienka Str., Kyiv, 04050, Ukraine.

Objective: Evaluation of structural features and metabolic/biochemical abnormalities of the bone tissue and relevant regulation patterns in children, residing in the radiologically contaminated territories (RCT).

Materials And Methods: Children (n = 148) aged 7 to 18 years old were involved in the study. Bone mineral density (BMD) is given in 3 grades according to the mean square deviation values, namely Grade I - standard (n = 75),Grade II - reduced (n = 45) and Grade III - very low one (n = 28).

Optimization of fermentation conditions and blending process of fairy bean in North Anhui Province.

AMB Express

December 2024

School of Biological Engineering, Huainan Normal University, Huainan, 232038, Anhui, People's Republic of China.

Functional fermentation strains were isolated and screened from traditional fairy beans in northern Anhui. Through technical identification, Bacillus subtilis SXD06 was determined to be the superior fermentation strain, while Wickerhamomyces anomalus YE006 was identified as the optimal aroma-producing yeast. Utilizing single-factor experiments and response surface optimization, a Central Composite Design fermentation and blending model was established.

The early stages of kidney crystal formation involve inflammation and hypoxia-induced cell injury; however, the role of the hypoxic response in kidney crystal formation remains unclear. This study investigated the effects of a prolyl hydroxylase domain inhibitor (roxadustat) on renal calcium oxalate (CaOx) crystal formation through in vitro and in vivo approaches. In the in vitro experiment, murine renal tubular cells (RTCs) were exposed to varying roxadustat concentrations and CaOx crystals.

Silver nanowire/gold nanosphere binary plasma-assembled membranes for sensitive SERS detection of homocysteine.

Mikrochim Acta

December 2024

School of Materials and Chemical Engineering, Fuzhou Institute of Oceanography, Minjiang University, Fuzhou, Fujian, 350108, China.

Silver nanowire (Ag NW)/gold nanosphere (Au NS) binary plasma films were prepared using plasma coupling between Ag NWs and Au NSs. The plasma films formed by combining these two noble metals showed better sensitivity for SERS detection with a minimum detection concentration of 10 M for R6G compared to pure Ag NWs or Au NSs. After rational optimisation of the substrate preparation process, the substrate showed good homogeneity, reproducibility and stability.
