Effective solubilization and single-step purification of Bacillus licheniformis alpha-amylase from insoluble aggregates.

A high level expression of thermostable alpha-amylase gene from Bacillus licheniformis in Escherichia coli was obtained. The recombinant enzyme was mainly produced in the form of insoluble aggregates. The enzyme was solubilized without using denaturing agents and purified to homogeneity in a single step by ion exchange chromatography. The enzyme was purified 138-fold with a final yield of 349 %; the specific activity of the purified enzyme was 1343 U/mg.

To investigate the bacterial community structure and physicochemical characteristics of different types of Daqu in the Binzhou region, this study employed traditional pure culture methods, high-throughput sequencing technology, and conventional physicochemical assays for analysis. Tomato is an important crop worldwide, but groundnut bud necrosis virus (GBNV) often hampers its growth. This study investigates the antiviral potential of bacterial endophytes, including CNEB54, CNEB4, CNEB26, and BAVE5 against GBNV, as well as their ability to enhance immunity and growth in tomato. All four bacterial isolates demonstrated a significant delay in GBNV symptom development 10 days post-inoculation, with disease incidence ranging from 18% to 36% compared to 84% in control.
