In poultry, prolactin (PRL) plays a key role in the regulation of incubation behavior, hormone secretion, and reproductive activities. However, previous in vitro studies have focused on the actions of PRL in ovarian follicles of poultry, relying on the use of exogenous or recombinant PRL, and the true role of PRL in regulating ovarian granulosa cell (GC) functions in poultry awaits a further investigation using endogenous native PRL. Therefore, in this study, we first isolated and purified recombinant goose PRL protein (rPRL) and native goose PRL protein (nPRL) using Ni-affinity chromatography and rabbit anti-rPRL antibodies-filled immunoaffinity chromatography, respectively.

Members of the Ets family of transcription factors are key regulators controlling prolactin (PRL) gene expression. Utilizing a transient transfection approach and the GH4 rat pituitary cell line, we have shown that Ets- 1 acts synergistically with the pituitary-specific POU homeodomain transcription factor, Pit-1, to mediate basal and Ras-induced regulation of the proximal (-425) rat PRL (rPRL) promoter. Although the transient transfection approach has provided important information regarding rPRL proximal promoter regulation, the role of Ets factors in the regulation of the intact, endogenous PRL promoter has not been explored.

Introduction: The purpose of this study was to induce hyperprolactinemia in adult male rats and determine its effect on the central and peripheral mediated neurogenic as well as myogenic mechanisms of erectile function/dysfunction in a rat model. The use of penile reflexes in awake animals was used to test the effect of both central and peripheral mechanisms. Electrical stimulation of the cavernous nervous was used to test peripheral neural function and intracorporal injection of papaverine was used to test the intactness of the corporal smooth muscles.
