Fluorescence spectroscopic methods to analyze drug-tubulin interactions.

Fluorescence spectroscopy has been extensively used to characterize ligand binding to tubulin and microtubules. The inherent advantages of fluorescence spectroscopic methods lie in their ease, sensitivity to local environmental changes, and ability to describe the protein-ligand interactions qualitatively as well as quantitatively in equilibrium conditions. In this chapter, we have described how fluorescence spectroscopy has been used to decipher molecular interaction between a wide variety of ligands and tubulin. Particularly, we have discussed its use to characterize the binding parameters of ligands that are known to bind to three important sites in tubulin namely the vinca domain, the colchicine binding site, and the taxol site. These are the sites where most of the microtubule-targeted anticancer agents bind to tubulin. An understanding of the interaction between tubulin and small molecule inhibitors can assist in understanding the cellular effects of these inhibitors. This will also help in developing molecules that have higher binding affinity to tubulin and can serve as potent anticancer agents.

A new twofold interpenetrated 3D metal-organic framework (MOF), namely, poly[[μ-aqua-diaqua{μ-2,2'-[terephthaloylbis(azanediyl)]diacetato}barium(II)] dihydrate], {[Ba(CHNO)(HO)]·2HO}, (I), has been assembled through a combination of the reaction of 2,2'-[terephthaloylbis(azanediyl)]diacetic acid (TPBA, HL) with barium hydroxide and crystallization at low temperature. In the crystal structure of (I), the nine-coordinated Ba ions are bridged by two μ-aqua ligands and two carboxylate μ-O atoms to form a 1D loop-like Ba-O chain, which, together with the other two coordinated water molecules and μ-carboxylate groups, produces a rod-like secondary building unit (SBU). The resultant 1D polynuclear SBUs are further extended into a 3D MOF via the terephthalamide moiety of the ligand as a spacer.

In this study, novel 2-styrylquinoline derivatives possessing a planar aromatic system and a flexible side chain with an amino substituent were designed and synthesized as DNA-intercalating antitumor agents. The cytotoxic activity of the synthesized compounds was evaluated against four cancer cell lines including MCF-7 (breast cancer cells), A549 (lung epithelial cancer cells), HCT116 (colon cancer cells) and normal cell line L929 (mouse fibroblast cell line). The results displayed that the anti-cancer activity of the target quinolines is sensitive to the lipophilic nature of the C-6 and C-7 quinoline substituents.
