A series of 10-position substituted nitrogenous heterocyclic aromatic group derivatives of SN-38 were prepared. Most of these compounds possessed lower cytotoxicities than CPT. Compound 13 revealed potent cytotoxicity similar to CPT, and compounds 17, 18, and 19 showed similar cytotoxic activity to topotecan. All of the pyridine salt derivatives (7-16) revealed comparable or superior topo I inhibitory activity in relation to CPT. Ethyl in the 7-position of these compounds can increase the cytotoxicity and inhibitory activity to topo I compared with corresponding pyridine salts CPT derivatives (7a-13a) and simultaneously maintain good water solubility. This result is consistent with the SAR of CPT.

Fluorescent probes for in vivo hypochlorous acid (HClO) imaging often face challenges of low selectivity and high cytotoxicity, largely due to poor analyte recognition and water-insoluble aromatic skeletons. To address this, we synthesized fluorescein hydrazide by introducing a spiro-lactam unit into fluorescein, which offers high emission intensity and molar absorption. The five-membered heterocycle in fluorescein hydrazide is selectively disrupted by HClO, enhancing the conjugated system and electron delocalization of the fluorophore, resulting in highly sensitive fluorescence detection of HClO.

N-heterocyclic carbene (NHC)-protected gold nanoclusters display high stability and high photoluminescence, making them well-suited for fluorescence imaging and photodynamic therapeutic applications. We report herein the synthesis of two bisNHC-protected Au nanoclusters with π-extended aromatic systems. Depending on the position of the π-extended aromatic system, changes to the structure of the ligand shell in the cluster are observed, with the ability to correlate increases in rigidity with increases in fluorescence quantum yield.
