Objective: to explore the value of denaturing high performance liquid chromatography (DHPLC) in detection of rpoB mutations in rifampin-resistant M. tuberculosis, in order to establish a convenient and rapid approach to screening rpoB gene mutations in M. tuberculosis.
Methods: rifampin resistance-determining region of rpoB gene in M. tuberculosis was amplified by PCR and further analyzed by DHPLC to screen mutations at optimized denaturation temperature (65.4 degrees C), which utilized heteroduplex formation between wild-type and mutated DNA strands to identify mutations. The PCR products from strains with different chromatographic profiles were sequenced further to evaluate the sensitivity and specificity.
Results: there were 46 M. tuberculosis strains including 42 rifampin resistance strains and 4 rifampin sensitive strains. From these strains, 15 different chromatographic profiles were produced by DHPLC. Combined with the results of gene sequencing, it was shown that strains with different chromatographic profiles had distinct mutations. Except D108 and D24 which had same chromatographic profiles but different genetic polymorphisms, all other strains showed consistent chromatographic profiles with genetic polymorphisms, i.e. if they had identical chromatographic profiles, their genetic polymorphism were the same; but if they had different chromatographic profiles, their genetic polymorphism were also different.
Conclusion: DHPLC is a simple, efficient, high-throughput and automatic method with high sensitivity and specificity, which may be useful in the rapid detection of rpoB gene mutation in M. tuberculosis.

Although proteins in snake venoms have been extensively studied and characterized, low-mass molecules remain relatively unexplored, mainly due to their low abundance, secondary role in envenomation, and some analytical technique limitations. However, these small molecules can provide new important data related to venom toxins' molecular structure, functions, and evolutionary relationships. This research aimed to characterize molecules below 10 kDa in the venoms of snakes from the Viperidae families (Bothrops, Agkistrodon, and Bitis) and compare two chromatographic approaches: reverse-phase chromatography (RP), a classic technique, and hydrophilic interaction liquid chromatography (HILIC), an alternative technique, both coupled with high-resolution mass spectrometry (HRMS).

Traditional packed beds in chromatography suffer from increased band broadening due to the random nature of packing, leading non-ideal fluid flow and channeling. To address these challenges, pillar array columns have been developed, offering improved performance over random packing thanks to their homogenous fluid profiles. The study aims to i) evaluate fluid dynamics and chromatographic performance across different PAC morphologies, ii) establish the influence of column morphology on performance, and iii) assess the correlation between chromatographic performance and hydrodynamic parameters.

The phytochemical profile of various plant species reveals that some compounds possess notable antioxidant and antimicrobial properties. In this study we investigated for the first time, the antioxidant activity (FRAP, DPPH and TAC), total phenolic contents and total flavonoid contents of Delile ex Godr flowers extracts (-hexane, ethyl acetate and methanol) as well as their antimicrobial activity. The results obtained showed that the methanol extract contained the highest content of total phenolics (346.
