The direct synthesis of six-fold symmetric hexaphenylbenzenes with multiple photochromic dithienylethene units via a cobalt-catalyzed cyclotrimerization is reported. This approach allows for six photochromic units to be held in proximity with a well-defined spatial separation without affecting the photochromic properties of each unit.

Herein, we present a hitherto unexplored efficient strategy for rapidly constructing structurally constrained and intriguing polycyclic frameworks with two adjacent quaternary centers. Remarkably, this becomes possible through palladium-catalyzed six-fold domino crossover annulations of simple 1,2-bis(2-bromoaryl)ethynes and 1,2-diarylethynes. Notably, this approach demonstrates the synthesis of both C-symmetric and unsymmetric polycyclic products.

Density functional theory computations were used to model the formation and rearrangement of the barbaralyl cation (CH ). Two highly delocalized minima were located for CH , one of symmetry and the other of symmetry, with the former having lower energy. Quantum chemistry-based NMR predictions affirm that the lower energy structure is the best match with experimental spectra.

Single-particle cryo-electron microscopy (cryo-EM) has been shown to be effective in defining the structure of macromolecules, including protein complexes. Complexes adopt different conformations and compositions to perform their biological functions. The severe acute respiratory syndrome-causing coronavirus 2 (SARS-CoV-2) papain-like protease (PL) and main protease (M) play an important role in viral replication events and are important targets for anti-coronavirus drug discovery. In search of these protease inhibitors, we screened a library of 1300 compounds using a fluorescence thermal shift assay (FTSA) and identified 53 hits that thermally stabilized or destabilized PL. The hit compounds structurally belonged to two classes of small molecules: thiazole derivatives and symmetrical disulfide compounds.
