Bioactive cage-like polycyclic compounds have attracted the attention of several research groups because of their unique appearance and their biological activities. Their structures were established on the basis of (1)H NMR and (13)C NMR spectroscopic data. The (1)H and (13)C signal assignments and most homonuclear hydrogen coupling constants were assigned by use of techniques such as 1D (1)H and (13)C NMR and 2D gCOSY, non-edited gHSQC and gHMBC. The gNOESY experiments proved the endo-stereochemistry.

In this study, polyamide/silica/sodium alginate (SA) composite (PA-Si-SA) was successfully prepared in one-step benzoxazine-isocyanide chemistry (BIC)/sol-gel process at room temperature. The chemical structure and fundamental properties of PA-Si-SA were characterized by FT-IR, solid-state C NMR, XPS, XRD, SEM, BET and TG, etc. The presence of anionic SA and diverse N, O-containing functional segments (amide, tertiary amine, alkyl/phenol -OH, Si-O-Si, and COO) in PA-Si-SA endows it synergistic complexation capability toward Pb and Cd.

Assignment of the N-terminal domain of mouse cGAS.

Biomol NMR Assign

January 2025

Department of Chemistry and Chemical Biology, TU Dortmund University, Dortmund, Germany.

Cyclic GMP-AMP synthase (cGAS) is a DNA-sensing enzyme that is a member of the nucleotidyltransferase (NTase) family and functions as a DNA sensor. The protein is comprised of a catalytic NTase core domain and an unstructured hypervariable N-terminal domain (NTD) that was reported to increase protein activity by providing an additional DNA-binding surface. We report nearly complete H, N, and C backbone chemical-shift assignments of mouse cGAS NTD (residues 5-146), obtained with a set of 3D and 4D solution NMR experiments.

Challenges of Investigating Compartmentalized Brain Energy Metabolism Using Nuclear Magnetic Resonance Spectroscopy in vivo.

Neurochem Res

January 2025

Department of Experimental Medical Science, Faculty of Medicine, Lund University, Lund, Sweden.

Brain function requires continuous energy supply. Thus, unraveling brain metabolic regulation is critical not only for our basic understanding of overall brain function, but also for the cellular basis of functional neuroimaging techniques. While it is known that brain energy metabolism is exquisitely compartmentalized between astrocytes and neurons, the metabolic and neuro-energetic basis of brain activity is far from fully understood.

Solution NMR studies of large systems are hampered by rapid signal decay. We hereby introduce ROCSY (relaxation-optimized total correlation spectroscopy), which maximizes transfer efficiency across J-coupling-connected spin networks by minimizing the amount of time magnetization spends in the transverse plane. Hard pulses are substituted into the Clean-CITY TOCSY pulse element first developed by Ernst and co-workers, allowing for longer delays in which magnetization is aligned along the z-axis.

Article Synopsis
  • Vinylene-linked Covalent Organic Frameworks (V-2D-COFs) are advanced materials known for their stable, crystalline structures and excellent optoelectronic properties, but their synthesis is limited due to challenges with C═C bonds.
  • Postsynthetic modification (PSM) offers a solution by allowing the introduction of functional groups into these frameworks, thus expanding their potential applications.
  • The study demonstrates a thiol-ene click reaction to successfully modify two COFs, enhancing their properties and structural versatility, which could lead to new applications in areas like hydrophilicity and proton conductivity.
