Label-free imaging through two-photon autofluorescence of NAD(P)H allows for nondestructive, high-resolution visualization of cellular activities in living systems. However, its application to thick tissues has been restricted by its limited penetration depth within 300 μm, largely due to light scattering. Here, we demonstrate that the imaging depth for NAD(P)H can be extended to more than 700 μm in living engineered human multicellular microtissues by adopting multimode fiber-based, low repetition rate, high peak power, three-photon excitation of NAD(P)H at 1100 nm.

While circular dichroism (CD) and optical activity (OA) are well established as optical effects used in characterization of chiral media, harmonic generation and multiphoton circular dichroism are increasingly seen as new, convenient ways of exploring chirality thanks to their operation in the near-infrared range of wavelengths. However, quantitative data about two-photon circular dichroism (2PCD) of organic and inorganic materials are scarce, and even less can be found about three-photon circular dichroism (3PCD). Here, we show that both 2PCD and 3PCD can be readily detected in chiral atomically precise gold nanoclusters via polarimetric Z-scan technique.

The nonlinear optical (NLO) properties of Ag-530 and Ag-610 NCs are compared with spherical silver NCs exhibiting plasmon peak at 420 nm (hence called Ag-420 NCs). Resonant excitation induces significant one-photon absorption (1PA) saturation for all NCs, with Ag-610 NCs displaying exceptional transparency.

Numerical simulations of the absorption bands of photoswitch --tetrafluoroazobenzene in DMSO solution under one-, two-, and three-photon absorption conditions combined with the analysis of the behavior of transition probability under distortion of planarity reveal many similarities between the mono- and triphoton spectroscopic behaviors with a two-photon spectrum being set apart. The position of the absorption peak for the studied nπ* and ππ* transitions appears shifted to lower energies (longer wavelengths) than the conventional estimate based on vertical excitation from the ground-state potential energy minimum.
