Background: Senile dementia (SD) is a deteriorative organic brain disorder and it comprises Alzheimer's disease (AD) as a major variant. SD is shown impairment of mental capacities whereas AD is degeneration of neurons. According to World Health Organization (WHO) report; more than 55 million peoples have dementia and it is raising 10 million new cases every year.

The glucose changes are consistent with brain thiamine (vitamin B1) deficiency. In animals, thiamine deficiency causes multiple AD-like changes including memory loss, neuron loss, brain inflammation, enhanced phosphorylation of tau, exaggerated plaque formation and elevated advanced glycation end products (AGE).

Given the desirability of a more acceptable administration route for prolonged AD treatment, we assessed the efficacy of HY209 via oral delivery. This study aims to elucidate the therapeutic potential of NuCerin, an oral formulation of HY209, in the aforementioned AD model, while simultaneously identifying potential blood biomarkers indicative of NuCerin's therapeutic action.

In these clinical studies, several plasma biomarkers assessments (Aβ42/40 ratio, p-tau181, GFAP, and p-tau217) showed improvements comparing lecanemab with placebo. Herein, we utilized modelling and simulations to evaluate the long-term effects of lecanemab on biomarkers of neurodegeneration in plasma.

Background: Research heavily suggests that brain-derived neurotrophic factor (BDNF), vital for neuronal growth and plasticity, and cholecystokinin (CCK), a satiety hormone that regulates BDNF levels, are altered in Alzheimer's Disease pathophysiology. Factors such as dysbiosis of gut microbiota and poor food habits may affect CCK and BDNF release and brain function. The objective is to evaluate the effects of dietary habits, gut microbiota, and exercise on BDNF and CCK release in Alzheimer's Disease patients.
