Background: Long-acting beta2-agonists (LABA) in combination with inhaled corticosteroids (ICS) are commonly used to treat asthma, however, some children lack response to the addition of LABA. This might be partially due to the presence of the Arg16Gly polymorphism, encoded by rs1042713 G>A in the ADRB2 gene. Carrying the A allele (Arg16) at this variant has been associated with an increased risk of exacerbations despite LABA treatment.

Genotype-driven asthma prescribing of inhaled corticosteroids and long-acting β2-agonist: A cost-effectiveness analysis.

Pediatr Pulmonol

October 2024

Research Group in Pharmacology and Toxicology, Department of Pharmacology and Toxicology, University of Antioquia, Medellín, Colombia.

Introduction: Predicting response to inhaled corticosteroids (ICSs) + long-acting β2-agonist (LABA) by previously detecting the presence of Arg16Gly ADRB2 genotype is a strategy that could reduce and optimize the management of asthmatic patients. There is a need for economic evaluations to facilitate the implementation of such tests. This research aims to evaluate the cost-effectiveness of Arg16Gly ADRB2 screening in children with asthma in Colombia.

Evaluation of ADRB2 and OATP2A1 genetic polymorphisms in Indian patients with primary open-angle glaucoma.

Pharmacogenet Genomics

January 2024

Department of Ocular Pharmacology and Pharmacy Division, Dr. Rajendra Prasad Centre for Ophthalmic Sciences, All India Institute of Medical Sciences, India Institute of Medical Sciences.

The emergence of adrenergic β2-receptor (ADRB2) blockers has revolutionized glaucoma treatment, while the discovery of prostaglandin analogs has further expanded therapeutic options. Organic anion transporting polypeptide 2A1 (OATP2A1/SLCO2A1) facilitates the corneal transport of topical prostaglandins into anterior segment of eye. Our study aims to elucidate the prevalence of genetic polymorphisms in the ADRB2 and OATP2A1 to address variations in therapeutic responses among glaucoma patients.

Cost-effectiveness of Arg16Gly in pharmacogenomic-guided treatment for pediatric asthma.

Expert Rev Pharmacoecon Outcomes Res

November 2023

International Peace Maternity and Child Health Hospital, School of Medicine, Shanghai JiaoTong University, Shanghai, China.

Objectives: To assess the cost-effectiveness of Arg16Gly pharmacogenomic testing compared with no Arg16Gly testing to guide the use of long-acting β receptor agonist (LABA) in asthma patients aged 1 to 5 years in China.

Methods: This economic evaluation developed a Markov model with four health states (no exacerbation, mild exacerbation, moderate-to-severe exacerbation, and death). Transition probabilities were estimated from the rate of exacerbations, the case-fatality rate of patients hospitalized for exacerbations, and natural mortality.

The ADBR2 gene has been studied for its possible relationship with the development and clinical course of chronic obstructive pulmonary disease (COPD), including response to beta-2 agonists, with existing data being contentious on the subject. So, the purpose of this study was to look into the potential impact of the arginine-16-glycine (Arg16Gly) polymorphism on the clinical course and drug utilization in COPD patients. Data show that patients with Arg16Arg have a lower number of hospital admissions for exacerbations (p=0.
