[Hydration of oocytes in bony fishes].

Ontogenez

Published: March 2010

Data are collected on the hydration of oocytes in bony fishes during maturation stimulated by gonadotropic or steroid hormones in vivo and in vitro. The reasons for hydration, its dynamics, and certain of the mechanisms ensuring income of water and ions into the oocyte are considered.

Electroneutral Na/Cl cotransport activity of zebrafish Slc12a10.1 expressed in oocytes.

School of Life Science and Technology, Tokyo Institute of Technology, Yokohama, Japan.

Na/Cl cotransporter 2 (Ncc2 or Slc12a10) is a membrane transport protein that belongs to the electroneutral cation-chloride cotransporter family. The Slc12a10 gene () is widely present in bony vertebrates but is deleted or pseudogenized in birds, some bony fishes, and most mammals. Slc12a10 is highly homologous to Ncc (Slc12a3 or Ncc1); however, there are only a few reports measuring the activity of Slc12a10.

Genetic adaptations of organisms living in extreme environments are fundamental to our understanding of where life can evolve. Water is the single limiting parameter in this regard, yet when released in the oceans, the single-celled eggs of marine bony fishes (teleosts) have no means of acquiring it. They are strongly hyposmotic to seawater and lack osmoregulatory systems.

College of Animal Science and Technology, Sichuan Agricultural University, Chengdu 611130, China.

Forkhead box H1 (FoxH1) is a sexually dimorphic gene in , , and , indicating that it is essential for gonadal development. In the present study, the molecular characteristics and potential function of FoxH1 and the activation of the promoter in vitro were evaluated in . The levels of in the ovaries were three times higher than those in the testes and were regulated by gonadotropins (Follicle-Stimulating Hormone and Human Chorionic Gonadotropin).

A new insulin-like growth factor (Igf) subtype 3 () has recently been found in the bony fish orange-spotted grouper (). However, the role of in the maturation of the ovary and sex differentiation in is currently unknown. We examined the ovarian localization and receptor binding of the novel ortholog Igf3 using qRT-PCR, and Western blotting, combined with in situ hybridization and immunohistochemistry methods.

Maternal effector genes (MEGs) encode maternal RNA and protein, accumulating in the cytoplasm of oocytes. During oocyte development, MEGs participate in oocyte meiosis and promote oocyte development. And MEGs can also regulate maternal transcriptome stability and promote maternal-zygotic transition (MTZ) in early embryonic development.
