Electron Transfer Dissociation of Oligonucleotide Cations.

Electron transfer dissociation (ETD) of multi-protonated 6 - 20-mer oligonucleotides and 12- and 14-mer duplexes is compared to collision activated dissociation (CAD). ETD causes efficient charge reduction of the multi-protonated oligonucleotides in addition to limited backbone cleavages to yield sequence ions of low abundance. Subsequent CAD of the charge-reduced oligonucleotides formed upon electron transfer, in a net process termed electron transfer collision activated dissociation (ETcaD), results in rich fragmentation in terms of w, a, z, and d products, with a marked decrease in the abundance of base loss ions and internal fragments. Complete sequencing was possible for nearly all oligonucleotides studied. ETcaD of an oligonucleotide duplex resulted in specific backbone cleavages, with conservation of weaker non-covalent bonds.

The interference of the expression of each of the genes involved in the synthesis of coenzyme Q (CoQ) in Drosophila melanogaster can help to understand the pathophysiology of CoQ-dependent mitochondrial diseases in humans. We have knocked-down all genes involved in the CoQ biosynthesis pathway at different temperatures to induce depletion of CoQ at different levels throughout the body and in a tissue-specific manner. The efficiency of the knockdowns was quantified by Q-RTPCR and determination of CoQ levels by HPLC-UV+ECD.

Sewer overflows are a potential source of emerging contaminants to urban waters, posing a threat to ecosystems and human health. Herein, the performance and mechanism of ferrate(Ⅵ) (Fe(Ⅵ))/peroxymonosulfate (PMS), Fe(Ⅵ)/peroxydisulfate (PDS), and Fe(Ⅵ)/percarbonate (SPC) for the degradation of ofloxacin (OFL) in overflows were comparatively investigated. These systems achieved efficient degradation of OFL and the removal of conventional pollutants.
