Background: Fragile X syndrome (FXS) is a leading known genetic cause of intellectual disability and autism spectrum disorders (ASD)-associated behaviors. A consistent and debilitating phenotype of FXS is auditory hypersensitivity that may lead to delayed language and high anxiety. Consistent with findings in FXS human studies, the mouse model of FXS, the Fmr1 knock out (KO) mouse, shows auditory hypersensitivity and temporal processing deficits.

Basic Science and Pathogenesis.

Alzheimers Dement

December 2024

Cleveland Clinic, Cleveland, OH, USA.

Background: Alzheimer's Disease (AD) risk variants APOE4 and TREM2-R47H have been shown to impact glial cell functions and transcriptional profiles. We hypothesize that TREM2-APOE may have synergistic effects in driving pathogenesis and disease progression of AD in a cell type-specific manner.

Methods: We investigated cell-type specific transcriptional changes associated with APOE4- and TREM2-R47H-carrier status.

Background: Maintaining gut microbial homeostasis is crucial for human health, as imbalances in the gut microbiota (GM) can lead to various diseases, including metabolic syndrome (MS), exacerbated by the use of antipsychotic medications such as olanzapine (OLZ). Understanding the role of the GM in OLZ-induced MS could lead to new therapeutic strategies. This study used metagenomic analysis to explore the impact of OLZ on the GM composition and examined how probiotics can mitigate its adverse effects in a rat model.

Serotonin Syndrome in Parkinson's Disease: Don't Get It Confused by a Tremor.

Cureus

November 2024

Intermediate Care Unit, Unidade Local de Saúde do Alto Minho, Viana do Castelo, PRT.

Serotonin syndrome (SS) is a potentially life-threatening condition caused by excessive serotonergic activity, often due to drug interactions. It classically manifests with autonomic and neuromuscular hyperactivity and by mental status changes that might include restlessness, delirium, and agitation. We present a case of a 76-year-old patient with Parkinson's disease with SS triggered by interaction between rasagiline and buspirone.

Acute respiratory distress syndrome (ARDS) results in decreased quality of life, including increased risk of pulmonary hypertension (PH). In animal models, ARDS can be induced by lipopolysaccharide (LPS), which can disrupt the pulmonary endothelium and epithelium and induce inflammation. We tested whether administration or treatment with LPS alters the reactivity of intrapulmonary arteries and airways to constrictors relevant to both ARDS and PH, using the precision cut lung slice (PCLS) technique.
