Development of fludarabine formulations in the treatment of chronic lymphocytic leukemia.

Fludarabine is an antineoplastic agent used in the treatment of hematological malignancies, particularly chronic lymphocytic leukemia (CLL) and indolent B-cell lymphoma. Because of its immunosuppressive effects, fludarabine has been added to reduced intensity conditioning regimens. The oral formulation of fludarabine has become widely available. Pharmacokinetic studies have shown that an oral dose of 40 mg/m(2)/d would provide systemic drug exposure similar to the standard intravenous (IV) dose of 25 mg/m(2)/d. The oral dose can be taken once daily without any dietary restrictions. Dose adjustments are mandatory in patients with renal impairment to avoid increased toxicity. Several noncomparative trials in previously untreated and treated patients with CLL have shown that treatment with the oral formulation demonstrates similar efficacy compared to historical control groups treated with the IV formulation. The tolerability profile of oral fludarabine seems similar to that of the IV formulation. Myelosuppression and infectious complications are the most frequently reported adverse events. Gastrointestinal toxicity is more frequent with the oral formulation, but is usually of mild or moderate severity. Although oral fludarabine makes treatment more convenient, health care workers must be aware of the compliance behavior of each patient.

Introduction: This study aimed to develop a deep learning-based method for interpreting magnetic resonance imaging (MRI) scans of temporomandibular joint (TMJ) anterior disc displacement (ADD) and to formulate an automated diagnostic system for clinical practice.

Methods: The deep learning models were utilized to identify regions of interest (ROI), segment TMJ structures including the articular disc, condyle, glenoid fossa, and articular tubercle, and classify TMJ ADD. The models employed Grad-CAM heatmaps and segmentation annotation diagrams for visual diagnostic predictions and were deployed for clinical application.

Purpose: This study aimed to develop a solid self-nanoemulsifying drug delivery system (SNEDDS) and surface-coated microspheres to improve the oral bioavailability of niclosamide.

Methods: A solubility screening study showed that liquid SNEDDS, prepared using an optimized volume ratio of corn oil, Cremophor RH40, and Tween 80 (20:24:56), formed nanoemulsions with the smallest droplet size. Niclosamide was incorporated into this liquid SNEDDS and spray-dried with calcium silicate to produce solid SNEDDS.

The purpose of this study was to investigate the effects of acetyl tributyl citrate (ATBC) on the mechanical properties, abrasion resistance, and cytotoxicity of a polyurethane-based 3D printing resin for mouthguard applications. The synthesized polycarbonate-based polyurethane acrylate was formulated into digital light processing printing resins with 40 wt% triethylene glycol dimethacrylate, and different percentage of ATBC were added for further characterizations. The mechanical properties and abrasion resistance, ATBC migration, and the cytotoxicity of the resins were evaluated.
