Fragment-Based Drug Discovery: Small Fragments, Big Impact - Success Stories of Approved Oncology Therapeutics.

Bioorg Chem

January 2025

Department of Pharmaceutical Chemistry, Manipal College of Pharmaceutical Sciences, Manipal, Academy of Higher Education, Manipal, Karnataka 576104, India.

Fragment-Based Drug Discovery (FBDD) has revolutionized drug discovery by overcoming the challenges of traditional methods like combinatorial chemistry and high-throughput screening (HTS). Leveraging small, low-molecular-weight fragments, FBDD achieves higher hit rates, reduced screening costs, and faster development timelines for clinically relevant drug candidates. This review explores FBDD's core principles, innovative methodologies, and its success in targeting diverse protein classes, including previously "undruggable" targets.

The photochemical inheritance of Eduardo Lissi and Juan Grotewold and the intersystem crossings with other inheritances.

Photochem Photobiol

January 2025

Departamento de Química, Universidad Nacional de Rio Cuarto, Río Cuarto, Argentina.

In 1963, Eduardo Lissi and Juan Grotewold started a chemical kinetics and photochemistry group at the School of Sciences at the University of Buenos Aires (Facultad de Ciencias Exactas y Naturales, FCEN, UBA). Political circumstances in Argentina and in Chile were a great determinant of the evolution, dispersion, and re-encounters of the group members. The initial graduate students in the group developed their own research groups working in various Countries and on a variety of projects.

The naturalistic paradigm and analytical methods present new approaches that are particularly suitable for research concentrating on narrative reading development. We analyzed fMRI data from 44 adults and 42 children engaged in story reading using time-locked inter-subject correlation (ISC), inter-subject representation similarity analysis (IS-RSA), and inter-subject functional correlation (ISFC). The ISC results indicated that for both children and adults, narrative reading recruited not only traditional reading areas but also regions that are sensitive to long-time-scale information, such as the medial prefrontal cortex and hippocampus, which increased involvement from children to adults.

Objectives: This research describes four aspects of the development of the Sense of Safety Theoretical Framework for whole person care: exploring the meaning of the phrase "sense of safety"-the whole person ; the range of human experience that impacts sense of safety-whole person ; the dynamics that build sense of safety-the healing ; and the personal and cross-disciplinary trauma-informed practitioner that facilitate sense of safety.

Methods: This qualitative participatory study was conducted in two phases. Researchers iteratively explored the concept of sense of safety using focus groups and semi-structured interviews.

This study presents a comprehensive workflow to detect low seismic amplitude gas fields in hydrocarbon exploration projects, focusing on the West Delta Deep Marine (WDDM) concession, offshore Egypt. The workflow integrates seismic spectral decomposition and machine learning algorithms to identify subtle anomalies, including low seismic amplitude gas sand and background amplitude water sand. Spectral decomposition helps delineate the fairway boundaries and structural features, while Amplitude Versus Offset (AVO) analysis is used to validate gas sand anomalies.
