Analysing the expression of protein kinase C eta in human hepatocellular carcinoma.

Aims: Protein kinase Cs (PKCs) play important roles in cell proliferation, differentiation, apoptosis, migration and tumorigenesis. In this report, we investigated the expression of PKCeta in human hepatocellular carcinoma (HCC) tissues and explored its role in the development of HCC.

Methods: We used real-time quantitative RT-PCR, mutation analysis, and immunohistochemical staining to analyse the expression of PKCeta in 50 pairs of human hepatocellular carcinoma (HCC) tissues.

Results: Expression of PKCeta was down-regulated in 82% of HCC tissues and the reduction of PKCeta was associated with poorer long-term survival of HCC patients.

Conclusion: Reduced expression of PKCeta may represent a molecular lesion in the development of more aggressive disease of HCC.

In this study, we aimed to elucidate the action mechanisms of propofol, particularly those underlying propofol-induced protein kinase C (PKC) translocation. Various PKCs fused with green fluorescent protein (PKC-GFP) or other GFP-fused proteins were expressed in HeLa cells, and their propofol-induced dynamics were observed using confocal laser scanning microscopy. Propofol-induced PKC activation in cells was estimated using the C kinase activity receptor (CKAR), an indicator of intracellular PKC activation.

Annexins (ANXs) comprise a family of calcium- and phospholipid-binding proteins and are implicated in the hepatitis C virus (HCV) life cycle. Here, we demonstrate a novel role of ANX5 in the HCV life cycle. Comparative analysis by quantitative PCR in human hepatoma cells revealed that ANX2, ANX4, and ANX5 were highly expressed among the ANX family proteins.
