Animal models of Parkinson's disease with dementia would greatly facilitate research into the underlying causes of this disorder. Here, we showed that bilateral infusion of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) into the substantia nigra pars compacta (SNc) of Wistar rats caused degeneration of nigrostriatal dopaminergic neurons, cell loss in the hippocampal CA1 area, as well as microglial activation and increase of interleukin-2 levels in several brain regions. In addition, increase of anxiety-like behavior and impairment of object recognition were observed in the MPTP-lesioned rats. These findings suggest that neuroinflammation may contribute to MPTP-induced neurodegeneration and behavioral deficits, which is suggested as an animal model of Parkinson's disease dementia.

Trans-active response DNA-binding protein-43 (TDP-43) is the major pathological protein in motor neuron disease and TDP-43 pathology has been described in the brains of up to 50% of patients with Alzheimer disease (AD). Hippocampal sclerosis of aging (HS-A), an age-related neuropathology characterized by severe neuronal loss and gliosis in CA1 and/or subiculum, is found in ∼80% of cases that are positive for phosphorylated TDP-43. HS-A is seen as a co-pathology in cases with AD, limbic-predominant age-related TDP-43 encephalopathy neuropathologic changes (LATE-NC), and frontotemporal degeneration.

Whisker deprivation at different stages of early development results in varied behavioral outcomes. However, there is a notable lack of systematic studies evaluating the specific effects of whisker deprivation from postnatal day 0 (P0) to P14 on adolescent behavioral performance in mice. To investigate these effects, C57BL/6J mice underwent whisker deprivation from P0 to P14 and were subsequently assessed at 5 weeks of age using a battery of tests: motor skills were evaluated using open field test; emotional behavior was evaluated using a series of anxiety- and depression-related behavioral tests; cognitive function was examined via novel location and object recognition tests; and social interactions were analyzed using three-chamber social interaction test.
