A 28-year-old male visited our hospital with complaint of chest pain. Clinical examination revealed a huge mediastinal mass which was diagnosed as non-seminomatous germ cell tumor. The patient underwent 5 cycles of chemotherapy (bleomycin, etoposide, and cisplatin) followed by resection of the tumor combined with left upper lobectomy. Final pathological diagnosis was germ cell tumor with somatic-type malignancy. While the prognosis of mediastinal germ cell tumor with somatic-type malignancy is known to be extremely poor, multimodality at an early stage is the key to successful treatment.

The H3K79 methyltransferase DOT1L is essential for multiple aspects of mammalian development where it has been shown to regulate gene expression. Here, by producing and integrating epigenomic and spike-in RNA-seq data, we decipher the molecular role of DOT1L during mouse spermatogenesis and show that it has opposite effects on gene expression depending on chromatin environment. On one hand, DOT1L represses autosomal genes that are devoid of H3K79me2 at their bodies and located in H3K27me3-rich/H3K27ac-poor environments.

In the presence of stressful environments, the SKN-1 cytoprotective transcription factor is activated to induce the expression of gene targets that can restore homeostasis. However, chronic activation of SKN-1 results in diminished health and a reduction of lifespan.
